基于中药量丰成分的药物发现研究

Drug Discovery Based on Naturally Abundant Ingredients

  • 摘要: 从中医药临床实践经验总结中,通过科学研究发现天然先导结构,进一步通过活性优化、成药性优化转化为创新药物,是实现中药现代化的有效途径。大量研究显示单味中药传统功效的物质基础通常是其量丰的特征性成分。通过对青蒿素、三氧化二砷、根皮苷等几种量丰特征性成分的活性发现、作用机制揭示、结构优化,最终开发成药物,总结了它们的成功经验,可为基于中药量丰成分的药物发现研究者提供可借鉴的研究思路。

     

    Abstract: From the clinical practices and summary of Chinese medicine, it has been proved that the discovery of lead compounds from natural medicine and followed by structural optimization toward innovative drugs is one of the most effective strategies for the modernization of Chinese medicine. A lot of research have exhibited that the chief pharmacological materials of single Chinese herb are always the naturally-abundant and structural featured constituent(s). In this perspective, we summarized the successful research stories of such constituents like artemisinin, arsenic trioxide and phlorizin, including discovery of the pharmacological activity, study of the molecular mechanism, structural optimization and development of innovative drugs. These experiences may contribute useful ideas for the researchers who are working in the field of drug discovery based on naturally abundant ingredients.
