藤梨根有效组分抑制胃癌BGC-823细胞增殖与迁移作用的研究

摘要: 目的 研究藤梨根有效组分对胃癌BGC-823细胞的体外增殖、迁移能力的影响,并初步探讨其相关机制。方法 利用不同浓度的藤梨根粗多糖与总三萜组合(熊果酸、齐墩果酸)处理细胞,MTT法检测细胞增殖活性,确定藤梨根有效组分最佳配比,划痕实验观察藤梨根有效组分对细胞运动能力的影响,Transwell小室模型研究其对细胞迁移能力的影响,基因芯片分析抑制BGC-823细胞增殖与迁移相关基因表达,qPCR检测miR-630基因的表达水平。结果 与空白对照组相比,当藤梨根粗多糖的浓度为0.5 g/L,熊果酸与齐墩果酸的浓度比为8∶1,各成分之间具有强协同作用,确定为藤梨根有效组分,其可显著抑制BGC-823细胞增殖与迁移,基因芯片分析和qPCR结果表明与其增加miR-630表达相关。结论 藤梨根有效组分能显著抑制胃癌BGC-823细胞的体外增殖和迁移能力,其机制可能与miR-630的表达水平增加有关。Abstract: OBJECTIVE To investigate the effects and mechanism of effective component of Kiwi root on inhibition of proliferation and migration of gastric cancer cell BGC-823. METHODS MTT to experiment the different concentration of Kiwi root polysaccharide with Kiwi root triterpene compounds, and to determine the best effective ratio, called Kiwi root effective component(KREC); The scratch adhesion test and Transwell assay were performed to determine the migration and invasion capacity of the cells. Gene chip were used to analyze the gene which was associated with the inhibition of proliferation and migration of gastric cancer cell BGC-823; Quantitative PCR were used to examine the expression of miR-630. RESULTS When the concentration of Kiwi root polysaccharide is 0.5 g/L, the concentration ratio of the ursolic acid to the oleanolic acid is 8:1,there is a strong synergy between the components, which can be called KREC and can restrain the proliferation and migration of gastric cancer cell BGC-823.The results of Gene chip and Quantitative PCR showed that the inhibiting ability was related to the increase of miR-630. CONCLUSION KREC have inhibitory effect on proliferation and migration of gastric cancer cell BGC-823, which mechanism can be related to increased expression levels of miR-630.
