糖可明对糖尿病视网膜病变中血-视网膜屏障的影响

摘要: 目的 探讨糖可明对糖尿病视网膜病变的影响。方法 用链脲佐菌素(STZ)复制大鼠糖尿病视网膜病变模型,观察糖可明对模型动物血液生化、血-视网膜屏障(Blood-retinal barrier,BRB)渗漏和视网膜VEGF、occludin表达的影响。结果 糖可明连续给药8周后,可明显降低GLU、TG和TC的含量(P<0.05~0.01);降低血-视网膜屏障的通透性;PCR结果显示,给药组大鼠视网膜occludin mRNA表达量显著增加(P<0.05~0.01);免疫组化实验发现糖可明可显著降低视网膜全层的VEGF表达(P<0.01)。结论 糖可明通过下调糖尿病大鼠视网膜VEGF的表达和增加occludin的表达,从而发挥其保护BRB、降低BRB通透性的作用,延缓糖尿病视网膜病变的进程。Abstract: OBJECTIVE To study the effect of Tangkeming (TKM) on blood-retinal barrier (BRB) in diabetic rats. METHODS Diabetic retinopathy rats were induced by STZ. The blood biochemical indexes were detected. Retinal vascular permeability was measured by Evans blue. The expression of occludin and VEGF in retina of diabetic rats were determined by qPCR and immunohistochemistry, respectively. RESULTS Administration of TKM to diabetic retinopathy rats significantly ameliorated the levels of GLU, TG and TC in serum. The Evans blue contents in dry retina tissue remarkedly reduced upon TKM treatment. The level of occludin mRNA was up-regulated and the expression of VEGF in retina was down-regulated by TKM in diabetic rats. CONCLUSION TKM protects the BRB by inhibiting expression of VEGF associated with occludin.
