三黄汤通过抑制脂肪组织间巨噬细胞浸润及炎症因子表达改善肥胖小鼠的胰岛素抵抗

Sanhuang Tang Improve Insulin Resistance in High Fat Diet Induced Obesity Mice by Inhibiting the Expression of Inflammatory Factors

  • 摘要: 目的 探讨三黄汤改善肥胖小鼠胰岛素抵抗与其抑制炎症因子表达的相关性。方法 C57BL/6雄性小鼠高脂饮食喂养12周诱导为肥胖鼠,随机分为模型组,三黄汤治疗组和二甲双胍阳性药组,治疗组分别以三黄汤和二甲双胍灌胃治疗2周;另设空白对照组小鼠给予正常饮食,模型组与空白对照组给予生理盐水。小鼠进行OGTT葡萄糖耐量实验,处死后取血清检测胰岛素、炎症因子表达;分离附睾脂肪组织以流式细胞术检测组织间浸润的CD8+ T细胞和巨噬细胞数量。结果 三黄汤降低肥胖小鼠的空腹血糖和空腹胰岛素,降低胰岛素抵抗指数,显著抑制脂肪组织间的巨噬细胞浸润,同时显著抑制血清TNF-α、IL-1β、MCP-1、Resistin的表达;三黄汤对脂肪组织间CD8+ T细胞数量无显著影响。结论 三黄汤可能通过抑制巨噬细胞浸润,抑制炎症因子表达改善肥胖小鼠的胰岛素抵抗。

     

    Abstract: OBJECTIVE To investigate the relationship between the inhibiting expression of inflammatory factors and improvement on insulin resistance treated by Sanhuang Tang. METHODS C57BL/6 male mice were respectively fed with high fat diet and normal diet for 12 weeks. HFD (High fat diet) mice were divided randomly to DIO model group, Sanhuang Tang treatment and metformin treatment group. Normal mice fed with normal diet were set as control group. The inflammatory factors expression in serum were examined by milliplex assay. The number of macrophages and CD8+ T cells in epididymis adipose tissue was examined by FACS. RESULTS Compared with DIO model group, Sanhuang Tang treatment significantly decreased the HOMA-IR factor, and slightly decreased fasting blood glucose and fasting insulin. At the same time, Sanhuang Tang significantly decrease the expression of TNF-α, IL-1β, MCP-1,resistin compared with model group. Sanhuang Tang significantly decrease the number of macrophages in epididymis adipose tissue, while did not significantly alter the number of CD8+ T cells. CONCLUSION Sanhuang Tang may improve insulin resistance induced by obesity in HFD mice by inhibiting macrophages infiltration and the expression of inflammatory factors.
