金振口服液治疗呼吸道合胞病毒感染引起咳嗽的作用机制研究

Study on the Mechanism of JZKFY Treatment of Cough Induced by Respiratory Syncytial Virus Infection

  • 摘要: 探讨金振口服液治疗呼吸道合胞病毒感染幼年豚鼠咳嗽的作用及其机制。方法 选取幼年豚鼠,随机分为空白组、模型组、磷酸苯丙哌林组、金振口服液(1.3、2.6g/kg)组。除空白组外,每鼠用106.29TCID50mL-1滴度的呼吸道合胞病毒(RSV)溶液滴鼻造模,空白组给予等体积无病毒DMEM溶液,次日起各给药组连续给药7d。第8天用枸橼酸引咳法检测豚鼠5min内咳嗽次数与咳嗽潜伏期,计数肺泡灌洗液中白细胞总数,ELISA法检测肺泡灌洗液中TNF-α、IL-4、IL-13含量,HE染色进行肺组织学检查,免疫组织化学染色法检测支气管中神经生长因子(NGF)、P物质(SP)、神经肽受体1(NK-1)的表达,Western blot法检测肺组织中NGF、NK-1的表达。结果 金振口服液能够抑制RSV感染导致的豚鼠咳嗽次数增多,下调肺泡灌洗液中白细胞总数及TNF-α、IL-4、IL-13的含量,改善肺组织的炎性浸润,降低支气管NGF、SP和NK-1及肺组织NGF和SP的表达。结论 金振口服液能改善RSV感染引起的病毒性咳嗽,其机制可能与减少气道炎性反应,降低神经源性介质,调节咳嗽敏感性有关。

     

    Abstract: OBJECTIVE To investigate the effect and mechanism of JZKFY on increaseed cough sensitivity of young guinea pigs after being infected with respiratory syncytial virus(RSV). METHODS Young guinea pigs were randomly divided into control group, model group, Benproperine group and JZKFY(1.3, 2.6g/kg) group. In addition to the control group, every guinea pig were treated with 106.29 TCID50 mL-1 titer of RSV solution, and the control group was given the same volume of virus-free DMEM solution. Every group were treated with medicine from the second day. The cough latency and frequency of guinea pigs were detected by citrate cough test at the 8th day; investigating the total number of leukocyte in alveolar lavage; the levels of TNF-α、IL-4、IL-13 were investigated by Elisa; the expression of nerve growth factor (NGF), substance P (SP), neurokinin1 (NK-1) in bronchus were detected by immunohistochemical staining; the expression of NGF and NK-1 in lung tissue were detected by Western blotting. RESULTS JZKFY could inhibit the increase of the number of cough caused by RSV infection, decrease the total number of leukocyte and the contents of TNF-α, IL-4 and IL-13 in bronchoalveolar lavage fluid, improve bronchial inflammation and reduce the expression of NGF,NK-1in bronchus and the expression of NGF and SP in lung tissue. CONCLUSION JZKDY can improve the inflammatory response inducted by RSV and have a therapeutic effect on cough after infection. The mechanism may be related to reduce the airway inflammatory reaction, decrease neurogenic medium and adjust cough sensitivity.
