“藻戟遂芫”与甘草合用对肠组织P糖蛋白转运功能的影响

Impact of "Zao-ji-sui-yuan" Combined with Glycyrrhizae on Rat Intestinal P-glycoprotein Transport Function in Vitro

  • 摘要: 目的 从离体肠组织P糖蛋白(P-gp)活性的角度,探讨中药“十八反”中“藻戟遂芫”与甘草合用产生禁忌的可能机理。方法 以罗丹明123(R123)作为指示剂,采用Ussing-chamber技术观察“藻戟遂芫”与甘草合用对大鼠离体回肠组织P-gp转运活性的影响。结果 甘草能明显促进R123的分泌(P<0.01),该作用可被P-gp抑制剂维拉帕米抑制;海藻、甘遂与芫花显著促进R123的分泌(P<0.05);海藻、大戟、芫花与甘草合用后R123分泌量与甘草组相比均明显下降(P<0.01);而甘遂与甘草合用后差异不显著。结论 甘草能明显诱导P-gp活性,与海藻、大戟、芫花合用后甘草对P-gp活性的诱导作用被拮抗,该作用可能导致药物毒性成分外排减少,肠组织损伤加剧,提示一些有毒中药与甘草合用后,肠组织P-gp活性的改变可能引起减效或增毒的相反效应。

     

    Abstract: OBJECTIVE The mechanism of Glycyrrhizae's incompatibility with "zao-ji-sui-yuan"(Sargassum, Euphorbia pekinensis, Euphorbia kansui and Daphne genkwa) was explored by assessing intestinal P-glycoprotein (P-gp) activity in vitro. METHODS Ussing-chamber technique was used to determine intestinal secretory function. Rhodamine123 (R123) was used as an indicator of P-gp activity. RESULTS Glycyrrhizae could significantly promote the secretion of R123. In the present of Sargassum, Euphorbia pekinensis and Daphne genkwa, the promotion effect of Glycyrrhizae was significantly suppressed. CONCLUSION P-gp activity can be significantly induced by Glycyrrhizae. The promotion effect of P-gp activity induced by Glycyrrhizae was antagonized by Sargassum, Euphorbia pekinensis and Daphne genkwa, which might lead to the reduction of toxic components pumping out and then aggravated intestinal tissue injury. These results indicated that when Glycyrrhizae combined with some toxic traditional Chinese medicine, change of the intestinal P-gp activity might cause the "contrary effects".
