RP-HPLC法测定山茱萸环烯醚萜总苷中马钱苷和莫诺苷含量

Simultaneous Determination of Loganin and Morroniside in Total Glucosides from Cornus officinalis by HPLC

  • 摘要: 目的 建立反相高效液相色谱法测定山茱萸环烯醚萜总苷中马钱苷和莫诺苷含量的方法,为完善山茱萸环烯醚萜总苷的质量评价提供依据。方法 色谱柱:Lichrosphere C18柱(4.6 mm×150 mm,5 μm);流动相:乙腈-水梯度洗脱(0~12 min,乙腈比例由10%增加到55%);流速1.0 mL/min;柱温30 ℃;检测波长240 nm。结果 马钱苷和莫诺苷分别在0.088~0.528 μg (r=0.999 8)和0.088~0.352 μg(r=0.999 5)之间范围内线性关系良好;平均回收率(n=6)分别为98.7%(RSD=1.3%)、98.6%(RSD=0.8%)。结论 所建立的方法简便、准确、分离效果好、重复性好,可以用于控制山茱萸环烯醚萜总苷的质量。

     

    Abstract: OBJECTIVE To set up a method to simultaneously determine loganin and morroniside in total iridoid glycosides of cornus officinalis by HPLC, and to provide the basis for the quality evaluation. METHODS The analysis was performed on a Lichrosphere C18 column and the mobile phase was composed of acetonitrile-water with a linear gradient elution. The flow rate was 1.0 mL/min, the temperature of column was 30 ℃, and the UV detection wave length was set at 240 nm. RESULTS Loganin and morroniside were linear in the ranges of 0.088~0.528 μg(r=0.999 8) and 0.088~0.352 μg(r=0.999 5), respectively. The average recoveries were 98.7%(RSD=1.3%)and 98.6%(RSD=0.8%). CONCLUSION The method of HPLC is accurate, easy to practice and reproducible, which can be used to control the quality of total iridoid glycosides of cornus officinalis.
