Abstract:
OBJECTIVE To establish a method for determining uridine, xanthine, thymine, hypoxanthine, inosine, guanosine, 2-deoxyguanosine, 2-deoxy inosine, thymidine, adenosine in the supernatant of Mactra veneriformis, Ruditapes philippinarum, and Meretrix meretrix Linnaeus by ethanol subsiding method. METHODS The xBridge C18 analytical column(4.6 mm×250 mm,5 μm) was used and the mobile phases were consisted of water and methanol with gradient elution. The flow rate was 0.5 mL/min, column temperature was at 25 ℃, and the wave length was 254 nm. RESULTS The supernatant of Mactra veneriformis, Ruditapes philippinarum and Meretrix meretrix Linnaeus by ethanol subsiding method contains uridine, xanthine, thymine, hypoxanthine, inosine, guanosine, 2-deoxyguanosine, 2-deoxy inosine, thymidine, and adenosine. CONCLUSION This method is easy to practice and accurate and can be used for determining the main nucleoside of supernatant by the ethanol subsiding method for clams.
