板蓝根乙酸乙酯部位抗病毒活性组分及相关化学成分研究

Antiviral Activity and Chemical Composition of Ethyl Acetate Extract of Radix Isatidis<\i>

  • 摘要: 目的 研究板蓝根乙酸乙酯部位的抗病毒活性组分及相关成分。方法 利用溶剂法及色谱法提取分离板蓝根乙酸乙酯部位各化学组分,MTT法体外筛选各组分抗单纯疱疹病毒1(HSV-1)活性,利用UPLC-Q-TOF-MS检测乙酸乙酯部位及各组分的化学成分,运用Markview软件及Excel软件进行各组分差异性成分分析,根据统计结果得到抗病毒相关的化学成分。结果 按照化学成分的相似性,利用色谱法将板蓝根乙酸乙酯部位分为16个组分,其中8个组分具有明显抗病毒活性。UPLC-Q-TOF-MS检测到乙酸乙酯部位中的49个化学成分,并明确了它们的化学结构,在各化学组分的分析检测及差异性分析中,发现了生物碱类、有机酸和氨基酸衍生物共12个化学成分与抗病毒活性相关。结论 板蓝根乙酸乙酯部位以生物碱、有机酸、核苷、黄酮、蒽醌及氨基酸类成分为主,其中生物碱、有机酸和氨基酸衍生物与抗病毒活性密切相关。

     

    Abstract: OBJECTIVE To investigate the antiviral activity and chemical composition of ethyl acetate extract of Radix isatidis. METHODS Each component of total ethyl acetate extracts was detected by solvent and chromatogram, the anti-HSV-1 activities of each component were detected by MTT assay in vitro, each component of total ethyl acetate extracts was analyzed by UPLC-Q-TOF-MS, and the data were analyzed by Markview and Excel software to find the difference among different components. According to statistics, the components related to antivirus activities were obtained. RESULTS According to the similarity of chemical composition, 16 components were extracted and separated from ethyl acetate extract of Radix isatidis by chromatography, and 8 components had antiviral activity. 49 chemical components were detected by UPLC-Q-TOF-MS from ethyl acetate extracts and their chemical structures were confirmed. Difference analysis of chemical components discovered 12 chemical components such as alkaloids, organic acids and amino acid derivatives associated with antiviral activity. CONCLUSION The ethyl acetate extract of Radix isatidis mainly contains alkaloids, organic acids, nucleosides, flavonoids, anthraquinones and amino acids, wherein the alkaloids, organic acids and amino acid derivatives are closely related to the antiviral activity.
