Abstract:
OBJECTIVE To observe the influence of puerarin on glucose and lipid metabolism in gestational diabetic mellitus(GDM) rats. METHODS High-fat feeding pregnant wistar rats were given streptozocin (STZ) 25 mg/kg intraperitoneally on the first day of pregnant. After injection for 72 hours, rats with FBG between 6.67-16.67 mmol/L were gestational diabetic models, which were randomly divided into two groups: GDM model group(MG, using 0.9% NaCl), puerain treated group(TG, using puerain 0.25 g/(kg·d), 6 rats in each group. And we selected 6 pregnant rats as normal gestation group(NG, using 0.9% NaCl). The rats were given drugs by intragastric administration until the offspring were born. The levels of FBG, insulin, serum lipid, TNF-α, IL-6, leptin and adiponection were observed at the age of 25 weeks. Leptin receptor insulin receptor, TNF-α, IL-6, adiponection and leptin mRNA expression in fat and liver tissues were detected by qPCR. RESULTS Compared with GDM model group, the levels of FBG, insulin, TG, TC, LDL-C, TNF-α, IL-6 and leptin of puerarin treated group were significantly decreased(

P<0.01). The levels of HDL-C and adiponection of puerarin treated group were significantly increased(
P<0.05,
P<0.01). The mRNA expression of IR in TG was higher than MG, and the mRNA expression of TNF-α and leptin in TG were decreased significantly (
P<0.05). CONCLUSION Puerarin can improve glucose and lipid metabolism in gestational diabetic rats, the mechanism may be related to the regulation of TNF-α, IL-6, leptin and adiponectin levels.
