艾灸调控IL-17对实验性RA大鼠血清RANKL和TGF-β含量的影响

Adjustment of Moxibustion on Serum Level of IL-17, RANKL and TGF-β in RA Rats

  • 摘要: 目的 探索艾灸治疗类风湿性关节炎(RA)抗炎的效应及预防骨破坏的IL-17信号转导机制。方法 雄性SD大鼠66只,随机分为空白组、模型组、空白艾灸组、艾灸组、IL-17过表达模型艾灸组、IL-17沉默模型艾灸组、IL-17过表达模型组、IL-17沉默模型组,前6组每组各9只,后2组每组各6只。大鼠右后足足垫部皮内注入弗氏完全佐剂0.15mL制作RA模型。造模成功后第2天进行慢病毒注射建立IL-17基因过表达、IL-17基因沉默模型。RA造模后第7天开始麦粒灸大鼠肾俞、足三里2穴各5壮,每日1次,治疗18d。观测计算大鼠足跖肿胀度、热痛域;采用ELISA法检测大鼠血清IL-17、RANKL、TGF-β含量。结果 造模后大鼠右后足容积显著高于空白组(P<0.01),热痛域值显著低于空白组(P<0.01);艾灸后大鼠右后足容积显著低于模型组(P<0.01),热痛域显著高于模型组(P<0.01)。模型组大鼠血清IL-17、RANKL显著高于空白组,血清TGF-β含量显著低于空白组(P<0.01);艾灸后艾灸组大鼠血清IL-17、RANKL显著低于模型组,血清TGF-β含量显著高于模型组(P<0.01)。IL-17过表达模型艾灸组大鼠RANKL含量显著高于艾灸组(P<0.01);IL-17过表达模型艾灸组大鼠TGF-β含量显著低于艾灸组(P<0.01)。IL-17沉默艾灸组大鼠RANKL含量显著低于艾灸组和空白组(P<0.01)。结论 艾灸可能通过调控IL-17,从而调节RA大鼠血清骨破坏因子RANKL以及骨保护因子TGF-β含量,起到抑制RA滑膜炎症后期骨破坏、保护骨组织的作用。

     

    Abstract: OBJECTIVE To explore the IL-17 signal transduction mechanism of anti-inflammatory effect of moxibustion on RA rats. METHODS 66 SD rats were randomly assigned to control, model, control+moxa, moxa, IL-17 overexpression+moxa, IL-17 knockdown+moxa, IL-17 overexpression and IL-17 knockdown groups. Complete freund's adjuvant were injected into the right paw of SD rats to establish RA model, and siRNAs were injected for IL-17 overexpression/knockdown models. After 7d, moxibustion treatments were performed for 18d. RESULTS The swelling degree of RA rats and the serum level of IL-17 and RANKL were significantly higher than control group(P<0.01), and the PWTL of RA rats,serum level of TGF-β were significantly lower than control group. After 18d of moxibustion treatment, the swelling degree of RA rats and the serum level of IL-17,RANKL were significantly lower than control group(P<0.01),and the PWTL of RA rats and serum level of TGF-β were significantly higher than control group. In addition, the serum level of RANKL in IL-17 knockdown+moxa rats were significantly higher than moxa group(P<0.01); the serum level of RANKL in IL-17 knockdown+moxa rats were significantly lower than moxa group and control group(P<0.01). CONCLUSION Moxibustion may adjust the serum level of RANKL and TGF-β by supressed the serum level of IL-17 therefore relieving RA in RA rats.
