铁自噬介导铁死亡在子宫内膜异位性疾病中的机制及中药干预策略

Mechanisms of Ferritinophagy-Mediated Ferroptosis in Endometriosis-Associated Diseases and Strategies for Traditional Chinese Medicine Intervention

  • 摘要: 子宫内膜异位性疾病发病机制复杂,临床治疗面临重大挑战。近年研究表明,病灶微环境中的铁代谢紊乱可通过激活铁自噬、释放游离铁,进而诱发铁依赖性的铁死亡,共同推动疾病的发生发展。中医药在调控铁代谢相关通路方面展现出独特优势,能够双向调节病灶细胞的铁死亡敏感性,改善病灶微环境。本文梳理了铁代谢紊乱驱动铁自噬介导的铁死亡的分子机制,并总结了中药复方及单体靶向相关通路治疗子宫内膜异位性疾病的研究进展,以期为子宫内膜异位性疾病的临床治疗提供新的理论依据和治疗策略。

     

    Abstract: The pathogenesis of endometriosis-associated diseases is complex, posing significant challenges for clinical treatment. Recent studies have demonstrated that iron metabolism dysregulation within the lesional microenvironment can activate ferritinophagy, leading to the release of free iron and subsequently inducing iron-dependent ferroptosis, which collectively drives disease initiation and progression. Traditional Chinese medicine (TCM) has shown unique advantages in modulating iron metabolism-related pathways, with the capacity to bidirectionally regulate the ferroptosis sensitivity of lesional cells and improve the lesional microenvironment. This review outlines the molecular mechanisms by which iron metabolism dysregulation drives ferritinophagy-mediated ferroptosis and summarizes the research progress on TCM formulas and active compounds that target relevant pathways in the treatment of endometriosis-associated diseases, aiming to provide a novel theoretical basis and therapeutic strategies for the clinical management of these diseases.
