电针对PSD大鼠行为学及神经营养因子和炎症因子的影响

Effect of Electroacupuncture on Regulation of Serum Inflammatory Factors and Nerve Cytokines and Nerve Function in PSD Rats

      目的  探讨电针疗法对脑卒中后抑郁症模型(PSD)大鼠行为学及血清炎症因子、海马组织神经营养因子的影响。
      方法  SD大鼠分为假手术组、卒中后抑郁组、电针组和氟西汀组,建立大鼠脑中动脉缺血(MCAO)模型,手术后进行慢性不可预测温和刺激(CUMS)4周,建立PSD模型同时进行电针治疗。治疗结束后进行行为学检测、TTC染色、HE染色、血清炎症因子、神经营养因子等方面检测。
      结果  与假手术组相比,MCAO手术后脑组织梗死面积明显增大;HE染色明显看出MCAO组脑组织损伤严重。经过PSD造模后,与假手术组相比,糖水消耗量明显减小,强迫游泳不动时间明显延长,水平与垂直活动距离无明显变化;炎症因子白细胞介素-1β(IL-1β)、白细胞介素-6(IL-6)、肿瘤坏死因子-α(TNF-α)含量升高;脑源性神经营养因子(BDNF)、胰岛素样生长因子1(IGF-1)、神经生长因子(NGF)含量下降;尼氏染色后明显看出海马神经元缺失。与卒中后抑郁组相比,电针组和氟西汀给药组糖水消耗量明显增加,强迫游泳不动时间明显缩短,水平与垂直活动距离无明显变化;炎症因子IL-1β、IL-6、TNF-α含量减小;神经营养因子BDNF、IGF-1、NGF含量增加;神经元明显增多。
      结论  电针疗法可以很好地改善PSD模型大鼠的抑郁症相关行为学变化,其机制可能与调节血清炎症因子,增加神经营养因子,促进神经保护有关。

     

    Abstract:
      OBJECTIVE  To investigate the effect of electroacupuncture on the regulation of serum inflammatory factors and neurocytokines and nerve function in PSD rats.
      METHODS  SD rats were divided into sham operation group, post-stroke depression group, electroacupuncture group and fluoxetine group. Middle cerebral artery occlusion(MCAO)model was established. After the operation, chronic unpredictable mild stimulation (CUMS) was performed for 4 weeks. The PSD model was established and electroacupuncture was performed simultaneously. After treatment, conduct behavioral testing, TTC staining, HE staining, serum inflammatory factors, neurotrophic.
      RESULTS  Compared with the sham operation group, the brain tissue infarct area was significantly increased after MCAO operation, and the neurological function defect was obvious; HE staining clearly showed that the MCAO group was seriously injured. After PSD modeling, compared with the sham operation group, the consumption of sucrose was significantly reduced, the time of forced swimming immobility was significantly prolonged, and there was no significant change in the horizontal and vertical movement distance; The content of inflammatory factors IL-1β, IL-6, TNF-α increased; the content of neurotrophic factors BDNF, IGF-1, and NGF decreased; the Nissl body was obviously missing after Nissl staining. Compared with the post-stroke depression group, the consumption of sucrose in the electroacupuncture group and the fluoxetine administration group was significantly increased, the time of forced swimming immobility was significantly shortened, and the horizontal and vertical movement distances did not change significantly, the content of inflammatory factors IL-1β, IL-6, TNF-α decreased, the content of neurotrophic factors BDNF, IGF-1, and NGF increased, which was statistically significant.Nissl body increased significantly.
      CONCLUSION  Electroacupuncture can improve the behavioral changes related to depression in PSD model rats, and its mechanism may be related to regulating serum inflammatory factors, increasing neurotrophic factors, and promoting neuroprotection.
