Mechanism of Qianyang Yuyin Granule on Renal Podocytes Injury in Spontaneously Hypertensive Rats

  •   OBJECTIVE  To investigate the therapeutic effect of Qianyang Yuyin Granule (QYYY) on hypertensive kidney injury.
      METHODS  spontaneously hypertensive rats (SHR) were chosen as the animal model to evaluate the QYYY therapeutic effect. HE and Masson staining were used to observe the kidney pathology in each treatment group. The expression of TRPC6 and podocyte damage-related proteins Desmin, Nephrin and Podocin was determined by Western blot assay. In addition, low, medium and high doses (QYYYL, QYYYM, QYYYH) of Qianyang Yuyin drug-containing serum were used to treat an in vitro rat podocyte injury model induced by angiotensin Ⅱ (Ang Ⅱ). Similarly, Western blot and immunofluorescence assays were used to determine the expression of TRPC6 and podocyte damage-related proteins.
      RESULTS  HE and Masson staining indicated that QYYY treatment markedly alleviated the kidney damage in SHR, which correlates with significant reduction of TRPC6 and Desmin, and induction of Nephrin and Podocin, as well as the improvement of Ang Ⅱ-induced podocyte injury.
      CONCLUSION  QYYY can ameliorate kidney and podocyte damage, and reduce hypertensive kidney damage.
