Simultaneous Determination of Loganin and Morroniside in Total Glucosides from Cornus officinalis by HPLC

Abstract
OBJECTIVE To set up a method to simultaneously determine loganin and morroniside in total iridoid glycosides of cornus officinalis by HPLC, and to provide the basis for the quality evaluation. METHODS The analysis was performed on a Lichrosphere C18 column and the mobile phase was composed of acetonitrile-water with a linear gradient elution. The flow rate was 1.0 mL/min, the temperature of column was 30 ℃, and the UV detection wave length was set at 240 nm. RESULTS Loganin and morroniside were linear in the ranges of 0.088~0.528 μg(r=0.999 8) and 0.088~0.352 μg(r=0.999 5), respectively. The average recoveries were 98.7%(RSD=1.3%)and 98.6%(RSD=0.8%). CONCLUSION The method of HPLC is accurate, easy to practice and reproducible, which can be used to control the quality of total iridoid glycosides of cornus officinalis.
