The Protective Effect of Radix Rehmanniae and Cornus Officinalis on HUVEC Stimulated by AGEs

Abstract
OBJECTIVE To detect the protective effect of Radix Rehmanniae and Cornus Officinalis on HUVEC stimulated by advanced glycation endoproducts (AGEs).METHODS The cultured HUVEC cells were randomly divided into normal, AGEs, Radix Rehmanniae, Cornus Officinalis, Radix Rehmanniae-Cornus Officinalis separated and combined decoction groups. Firstly, adding 0.125 g/L,0.062 5 g/L,0.031 25 g/L final concentration drugs in advance for 1 h. Secondly, using 200 mg/L AGEs as stimulus to continue culturing for 24 h. Thirdly, using CCK-8 to measure and calculate the protection rate of drugs on HUVEC cells stimulated by AGEs. Last, measuring SOD, MDA, NO, LDH, TNF-α, IL-1β by ELISA method. RESULTS Different density of Radix Rehmanniae, Cornus Officinalis, and Radix Rehmanniae combining Cornus Officinalis all have protective effect on HUVEC cells stimulated by AGEs. The SOD, NO decreased (P<0.05~0.01), MDA, LDH, TNF-α, IL-1β increased (P<0.05~0.01) in the supernate of AGEs group. Medication administration groups can increase SOD, NO, decrease MDA, LDH, TNF-α, IL-1β, and have a significant difference compared to AGEs group (P<0.05~0.01).CONCLUSION Radix Rehmanniae, Cornus Officinalis, and Radix Rehmanniae combining Cornus Officinalis can protect vascular endothelial cells by increasing production of NO, inhibiting oxidative stress, and regulating emiocytosis. Especially the Radix Rehmanniae combining Cornus Officinalis group has a more obvious effect.
