Abstract
OBJECTIVE To study the anti-proliferative activity and mechanism of Xiao'ai Jiedu decoction against breast cancer cell lines MDA-MB-231. METHODS The cell morphology was observed by microscope. MTT method was used to investigate the inhibitory rate of Xiao'ai Jiedu decoction against breast cancer cell lines MDA-MB-231. Flow cytometry analyzed the cell cycle distribution. Western blot detected the expression of Cyclin B1 and CDK1 and the expression of upstream signaling molecules P53, P21 in PI3K/AKT signaling pathway. RESULTS Compared with the control group, MDA-MB-231 cells of Xiao'ai Jiedu decoction group showed number-decreased, loosely arranged, morphology-smaller and rounder. Xiao'ai Jiedu decoction inhibited the proliferation of MDA-MB-231 cells(P<0.05, P<0.01). Xiao'ai Jiedu decoction down-regulated the expression of Cyclin B1 and CDK1, inhibited the PI3K/AKT signaling pathway, and up-regulated the expression of P53 and P21(P<0.05, P<0.01). CONCLUSION Xiao'ai Jiedu decoction can inhibit the proliferation of MDA-MB-231 breast cancer cells, perhaps related to inhibiting the PI3K/AKT signaling pathway.
