Simultaneous Determination of Flavones and Scutebarbatines in Scutellariae barbatae by HPLC

  • OBJECTIVE To establish HPLC methods for simultaneous determination of luteolin, apigenin, wogonin, scutebarbatine A and scutebarbatine B.METHODS The analytical column was Phenomenex Luna C18(250 mm×4.60 mm, 5 μm). The mobile phase was composed of acetonitrile and water with gradient elution. The flow rate was 1.0 mL/min; The column temperature was 30 ℃. Luteolin and apigenin were detected at 355 nm; wogonin, scutebarbatine A and scutebarbatine B were detected at 264 nm.RESULTS The ranges of calibration curve of luteolin, apigenin, wogonin, scutebarbatine A and scutebarbatine B were 0.037 4~2.40 μg、0.031 2~2.00 μg、0.031 2~2.00 μg、0.006 2~0.40 μg and 0.006 2~0.40 μg, respectively. Also, the intra-batch and inter-batch variations and the average recoveries were all meet the requirement of Chinese Pharmacopoeia.CONCLUSION The method is rapid and simple with a good reproducibility and it can be used for the quality control of herba Scutellariae barbatae.
