紫草素对乙酰胆碱-氯化钙混合液诱导的房颤大鼠的保护作用研究

Protective Effect of Shikonin on Ach-CaCl2-Induced Atrial Fibrillation in Rats

  • 摘要: 目的 探索紫草素对乙酰胆碱-氯化钙(Ach-CaCl2)诱导的房颤大鼠的保护作用及可能机制。方法 采用雄性SD大鼠尾静脉注射Ach-CaCl2混合液建立大鼠房颤模型,分为正常对照组、模型组与紫草素组。其中,紫草素组预先1周灌胃给予紫草素4 mg/(kg·d),再与模型组同时给予Ach-CaCl2混合液造模,采用心电图检测、心脏彩超观察、Masson染色、ELISA法、Western blot等方法评价紫草素对房颤大鼠的保护作用。结果 心电图结果显示紫草素延长了大鼠房颤诱发时间,彩超结果显示紫草素改善房颤大鼠心脏扩大情况,ELISA结果显示紫草素降低了房颤大鼠血清炎症因子IL-6和TNF-α水平,Western blot法同样表明紫草素抑制了房颤大鼠心房组织的IL-6和TNF-α的蛋白表达。结论 紫草素对房颤大鼠具有保护作用。

     

    Abstract: OBJECTIVE To evaluate the potential therapeutic effects and mechanism of shikonin on atrial fibrillation in vivo. METHODS Male Sprague-Dawley rats were injected with Ach-CaCl2 mixture to make atrial fibrillation model. They were divided into blank group, model group and shikonin group. 4 mg/(kg·d) Shikonin was given to the shikonin group by one-week gavage in advance, Then given Ach-CaCl2 mixture at the same time. The effects of shikonin on atrial fibrillation in rats were evaluated by electrocardiogram, color Doppler ultrasound, Masson staining, ELISA and Western blot. RESULTS Electrocardiogram showed that shikonin prolonged the induction time of atrial fibrillation; color ultrasonographic results showed that shikonin improved heart enlargement in rats with atrial fibrillation; shikonin inhibited serum inflammatory factors IL-6 and TNF-α in rats with atrial fibrillation; Western blot also showed that shikonin inhibited the expression of IL-6 and TNF-α in atrial tissue of atrial fibrillation rats. CONCLUSION Shikonin shows a protective effect on a rat model of atrial fibrillation.
