加味泽泻方中挥发油环糊精包合物的制备及其对透皮扩散的影响研究

Preparation of Cyclodextrin Inclusion Compound of Volatile Oil in Jiawei Zexie Fang and Study on its Effect of Transdermal Diffusion

  • 摘要: 目的 制备加味泽泻方中挥发油的环糊精包合物,并对其透皮扩散的影响进行研究。方法 分别采用饱和水溶液法和研磨法制备挥发油的环糊精包合物,用HPLC法测定其包合率及载药量,并对包合物的外观形态、稳定性、体外释放度及离体透皮实验进行研究。结果 离体透皮实验证明,包合物24h药物累积释放量均低于挥发油/可溶性淀粉对照组,累积释放量排序为挥发油/HP-β-环糊精包合物>挥发油/DM-β-环糊精包合物>挥发油/β-环糊精包合物,与包合物的稳定性研究排序相反。结论 加味泽泻方中挥发油/环糊精包合物延缓挥发油释放。

     

    Abstract: OBJECTIVE To prepare cyclodextrin inclusion compound of volatile oil in Jiawei Zexie Fang and study its transdermal diffusion effect. METHODS Use saturated aqueous solution method and grinding method to prepare cyclodextrin inclusion compound of volatile oil respectively. The inclusion rate and drug loading were determined by HPLC. The appearance, stability, in vitro release and in vitro transdermal experiments were investigated.RESULTS The vitro transdermal delivery experiment showed that drug cumulative release of inclusion compound were lower than volatile oil/soluble starch control group in 24h and the order of drug release was as following, volatile oil/HP-β-cyclodextrin inclusion complex>volatile oil/DM-β-cyclodextrin inclusion complex>volatile oil/β-cyclodextrin inclusion complex, which was contrary to the order of the inclusion compound stability study.CONCLUSION The volatile oil/cyclodextrin inclusion compound can delay the release of volatile oil extracted from Jiawei Zexie Fang.
