2022, 38(2): 93-97. doi: 10.14148/j.issn.1672-0482.2022.0093
Abstract:
Professor Zhou Zhongying's experience in the differentiation treatment of tumors was reviewed from the pathogenesis theory of cancer toxin, and the dialectical relationship between "supporting the healthy energy, tonifying the deficiency" and "fighting against cancer and detoxifying" was elaborated. We proposed a tumor treating strategy with fighting against cancer and detoxifying as the core treatment, supplemented by strengthening the spleen and stomach to support the healthy energy. According to the pathogenic characteristics that the healthy energy is prone to cancer toxin, it is suggested that special attention should be paid to the application of the treatment method and related formulas to benefit qi and nourish yin in supporting the healthy energy combined with tonifying the deficiency, which can provide guidance and reference for clinical anti-tumor treatment.

2022, 38(2): 98-102. doi: 10.14148/j.issn.1672-0482.2022.0098
Abstract:
Through compiling and studying the related literature of Jinshui Liujun Decoction, we find that this formula is created based on Erchen Decoction and Zhenyuan Yin. Especially in drying dampness and dissolving phlegm aspects, rehmanniae radix praeparata and angelicae sinensis radix are added into it to warm and supplement genuine yin for treating deficiency-cold, highlighting the formulation idea of tonifying original qi, as well as warming the spleen and strengthening the kidney. In addition, this formula was used by Zhang Jingyue for treating 19 kinds of diseases and syndromes, and it was also widely used by later generations of physicians with flexible modifications. Therefore, the study on the medicine compatibility characteristics and the formulation idea of Jinshui Liujun Decoction is of great practical value and guidance for the later generations to understand Zhang Jingyue's harmonization treatment method, to study his academic idea of warming and supplementing genuine yin, and to broaden the clinical ideas of treating phlegm along with panting syndrome.

Abstract:
OBJECTIVE  To observe the effects of Huangqi-Ezhu-Chonglou combination (HEC) on the permeability and tight junction of human umbilical vein endothelial cells (HUVEC) and to explore the mechanism of HEC on inhibiting the hematogenous metastasis of colon cancer cells (HCT116).  METHODS  Control serum and HEC-medicated serum were prepared. The co-culture system of HUVEC-HCT116 was constructed and divided into single culture control group, co-culture model group and HEC low, medium and high dose (2.1, 4.2, 8.4 g ·kg-1) groups. In addition, HUVEC single culture control group was set up. Cell proliferation was detected by CCK-8 method; FITC-dextran method and transwell method were used to detect the permeability of HUVEC and the number of cross endothelial migration of HCT116 cells; The distribution of ZO-1 protein in HUVEC was detected by immunofluorescence; The protein expressions of RhoA, ROCK and ZO-1 were detected by Western blot.  RESULTS  The proliferation of HUVEC was inhibited after 48 h of treatment with different doses of HEC medicated serum (P < 0.01). Compared with the single culture control group, FITC-dextran penetration and the number of transendothelial migration of HCT116 in the co-culture model group significantly increased (P < 0.01), the expression of ZO-1 was down-regulated (P < 0.01), and the expressions of RhoA and ROCK were up-regulated(P < 0.01); Compared with the co-culture model group, the FITC-dextran penetration and the number of transendothelial migration of HCT116 in the HEC group significantly decreased (P < 0.05), the expression of ZO-1 protein was up-regulated (P < 0.05), and the expressions of RhoA and ROCK were down-regulated in a dose-dependent manner (P < 0.05).  CONCLUSION  HEC may regulate the expression of tight junction associated proteins ZO-1 in HUVEC by inhibiting RhoA/ROCK pathway, reduce vascular endothelial permeability, and then inhibit colon cancer hematogenous metastasis.

Abstract:
OBJECTIVE  To explore the potential mechanism of Wenshen Yanggan Decoction on protecting dopaminergic neurons and delaying the process of Parkinson's disease (PD).  METHODS  Fifty mice were randomly divided into 5 groups (n=10): control group, model group, Wenshen Yanggan Decoction high dose group, Wenshen Yanggan Decoction low dose group and amantadine group. Except for the control group, mice were injected intraperitoneally with 30 mg · kg-1 of 1-methyl-4-phenyl-1, 2, 3, 6-tetrahydropyridine (MPTP) twice a week for 4 consecutive weeks to establish a Parkinson's disease (PD) mouse model. The mice were administrated with Wenshen Yanggan Decoction by gavage once a day for 3 weeks. The behavioral effects of mice were observed. Immunohistochemistry and Nissl staining were used to detect the number of DA neurons. The contents of DA, 3, 4-dihydroxyphenylacetic acid (DOPAC) and homovanillic acid (HVA) in pathological tissues were measured by HPLC. Colorimetric method was used to determine the activities of ATPase and mitochondrial respiratory chain ComplexⅠ in the substantia nigra. The protein expression levels of PINK1, Parkin, VDAC1, LC3Ⅱ/Ⅰ, and P62 were detected by Western blot.  RESULTS  The behavioral tests showed that compared with the model group, Wenshen Yanggan Decoction significantly improved the muscle strength and exercise balance of mice (P < 0.01), and prolonged the duration of mice on the roller (P < 0.01). The Results of the immunohistochemistry demonstrated that a large number of TH immunoreactive cells and obvious cell protrusions were observed in Wenshen Yanggan Decoction high dose group, which was consistent with the Results of the Nissl staining assay. In addition, Wenshen Yanggan Decoction significantly increased the levels of DA and HVA (P < 0.05, P < 0.01) in the striatum. Furthermore, treatment with the high dose Wenshen Yanggan Decoction profoundly promoted the activities of ATPase and ComplexⅠ(P < 0.01), up-regulated the protein levels of PINK1 (P < 0.05), Parkin (P < 0.01) and LC3Ⅱ/Ⅰ (P < 0.01), and inhibited P62 protein expression (P < 0.01) in MPTP-induced mice.  CONCLUSION  Wenshen Yanggan Decoction can improve mitochondrial function, and provide neuroprotective effect in PD by activating PINK1/Parkin mediated mitochondrial autophagy pathway.

Abstract:
OBJECTIVE  To investigate the hair growth-promoting activity and the possible mechanism of pine massoniana needle extract (PMNE) on mice by using the dihydrotestosterone (DHT) induced androgenetic alopecia (AGA) model.  METHODS  48 C57BL/6 mice were randomly divided into 6 groups: control group without any treatment, DHT group (30 mg ·d-1 DHT only), procyanidin group (DHT+procyanidin B2 5 mg ·kg-1), PMNE low dose group (DHT+PMNE 4 mg ·kg-1), PMNE medium dose group (DHT+PMNE 8 mg ·kg-1) and PMNE high dose group (DHT+PMNE 12 mg ·kg-1). PMNE and procyanidin B2 were given orally every day. The hair growth of mice was observed and evaluated at d7, d14, d21 and d28 after administration. The circle skins of all mice were harvested at the same depilated aera after 28 days. The hair from these skins was weighed. Some skin tissue section was obtained for HE staining, and the number of hair follicles were calculated. Some skin tissues were obtained to detect the contents of ROS and MDA and the mRNA and protein levels of Keap1, Nrf2, NQO1, HO-1 and TGF-β1 by qPCR and Western blot.  RESULTS  PMNE promoted the hair growth of AGA mice and showed a dose-dependent effect. PMNE of 12 mg ·kg-1 could alleviate the DHT-induced AGA in mice and achieve a similar effect to procyanidin B2. PMNE reduced the contents of ROS and MDA, promoted the mRNA and protein expressions of Nrf2, NQO1, HO-1 and reduced the mRNA and protein expressions of Keap1 and TGF-β1.  CONCLUSIONS  PMNE can promote the hair growth of AGA mice by regulating the Nrf2-ARE signaling pathway.

2022, 38(2): 136-146. doi: 10.14148/j.issn.1672-0482.2022.0136
Abstract:
OBJECTIVE  To explore the anti-tumor mechanism of Jianpi Jiedu Decoction (JPJDD) based on network pharmacology and molecular docking.  METHODS  TCMSP, TCMID and ETCM databases were used to predict the active ingredients and the targets of JPJDD, and GeneCards database was used to search tumor related targets and match the targets above. The common targets were imported into Cytoscape 3.7.2 to construct the PPI network and select the key targets by network topology analysis. GO enrichment analysis and KEGG pathway enrichment analysis were performed by R 3.6.3 software. Finally the Autodock 2.4 was used for molecular docking to predict the binding degree of the active ingredients to target genes. In addition, the key targets were verified by cell experiments.  RESULTS  24 kinds of active ingredients and 86 tumor related targets were identified, including the key targets of PTGS2, HSP90AA1 and PRSS1, which were obtained by PPI network topology analysis. The molecular docking showed that all of the key targets had good binding activity with quercetin and kaempferol. GO enrichment analysis and KEGG pathway enrichment analysis suggested that JPJDD may play the role of anti-tumor through PI3K/Akt signal pathway and MAPK signal pathway. The results showed that JPJDD inhibited both the protein and mRNA expressions of PTGS2 and p38MAPK, but after silencing PTGS2 gene, the expression of downstream gene p38MAPK and the effect on colorectal cancer cells metastasis had no obvious response to JPJDD.  CONCLUSION  JPJDD can inhibit the metastasis of colorectal cancer via PTGS2 mediated p38MAPK signaling pathway.
