New System Construction for Syndrome Differentiation of Traditional Chinese Medicine Pathogenesis

Abstract:
With the significant changes of clinical problems faced by traditional Chinese medicine, we should attach importance to new theoretical innovation on the basis of inheritance and clinical practice. The formation of syndrome differentiation system originates from Treatise on Febrile Diseases written by Zhang Zhongjing. Although different methods of syndrome differentiation have their own applicable scopes and characteristics, the generality of them lies in grasping the pathogenesis. Professor Zhou Zhongying believes that pathogenesis analysis should be put on the top priority for syndrome differentiation. A new system of syndrome differentiation and treatment should be constructed with pathogenesis as the core and multiple syndrome differentiation as a whole, so as to refine a more practical, flexible and comprehensive method for syndrome differentiation, which will not only activate the syndrome differentiation, but also make it easier to master. The basic elements of syndrome differentiation concerning pathogenesis include pathological factors, nature of disease, location of disease and condition of disease, all constituting the syndrome factors of diagnosis of syndrome differentiation. We should grasp the structure of pathogenesis in the application of syndrome differentiation of pathogenesis, clarify the complexity of compound pathogenesis and distinguish the identity of pathogenesis from its difference. The diagnosis and treatment scheme should be made with disease pathogenesis as the main part in syndrome differentiation of pathogenesis. And the syndrome elements should be taken as the contents, which contain syndrome differentiation, nature of disease, location of disease, condition of disease, evolution of disease as well as its treatment, prescription samples, modification and clinical essentials.
Exploration on Compatibility of Herbs for Nourishing Yin in Formulas of Kidney-yang Tonification

Abstract:
To explore the compatibility characteristics of herbs used for nourishing yin in formulas of kidney-yang tonification. It is noticed that the appearance frequency of such herbs for yin nourishment as Radix Rehmanniae Praeparata, Rhizoma Dioscoreae, Fructus Corni and Fructus Lycii comes out on the top. Either the dosage ratio or the number of herbs of yin nourishment in the modern formulas for tonifying kidney yang is evidently increased than that of the ancient formulas, with the application scope being expanded. Hence, it is believed that compatibility of herbs for nourishing yin in formulas of kidney-yang tonification has become a major compatibility form in the clinical practice. When deficiency of kidney essence is obvious, the dosage of herbs for nourishing yin should be increased. While when the internal generation of deficient cold takes a dominant place, the dosage should be decreased, which is able to provide a reference for the reasonable application of formulas for kidney-yang tonification in clinical practice.
Exploration on Chapter of Jueyin Diseases of Treatise on Febrile Diseases

Abstract:
There have always been many controversies in chapter of jueyin diseases of Treatise on Febrile Diseases, including three aspects, namely, the doubt about main syndrome of jueyin diseases, the dispute of Wumei Pill as main prescription of jueyin diseases and the affiliation of articles on syncope, diarrhea, vomiting and hiccup. On the basis of previous studies, further investigation and discussion were made on these three issues. The pathogenesis of advance and retreat of cold-heat goes throughout the chapter of jueyin diseases, whose meaning lies in not only revealing the outcome of six-meridian diseases, but also playing a vital role in the research of various integrative cold and heat prescriptions of Zhang Zhongjing.

Abstract:
"Symptom set" was changed into "combined set of syndrome factors" by exploring the mapping relationship between symptom and syndrome factors, among which the ones meeting the definition of pathogenesis were chosen to further form the "pathogenesis set" under the ISOR combination principle, so as to build a new syndrome factor system of "symptom collection-syndrome factor collection-pathogenesis collection", which is not only beneficial to perfect the syndrome factor system, but also a new thought for applying mathematics and computer science to Chinese medicinal research.
Study on Serum Proteomics of Rheumatism with Damp-heat Syndrome Based on iTRAQ Labeling Method Combined with Bioinformatics Technology

Abstract:
OBJECTIVE To investigate the serum proteomics of rheumatism with damp-heat syndrome by using iTRAQ labeling method together with bioinformatics technology. METHODS 20 serum samples respectively from ankylosing spondylitis with damp-heat syndrome group, arthrolithiasis with damp-heat syndrome group and normal control group were collected for proteomics analysis through iTRAQ labeling and liquid chromatography. Such network database platforms as DAVID, UniProtKB/Swiss Prot and IPA were resorted to perform bioinformatics analysis of differential protein in rheumatic diseases with damp-heat syndromes and verification would be done to the significant differential proteins. RESULTS Fifteen differential proteins of dampness-heat syndrome were obtained, with twelve being the up-regulated expression proteins and three being the down-regulated expression proteins. The five typical biological signaling pathways of most significant meaning related to differential protein expression were obtained through analysis. These differential protein functions and bioinformatic analysis were proved to be related to the acute phase responses and activities of diseases. Verification test results were consistent with those of iTRAQ. CONCLUSION This provides evidences for further study on molecular mechanism of rheumatic diseases with damp-heat syndromes as well as the candidate diagnostic markers.
Clinical Study on Modified Qingying Decoction in Treating Endotoxemia in Cirrhosis due to Hepatitis B of Toxic Heat and Blood Stasis Syndrome

Abstract:
OBJECTIVE To study the effects of modified Qingying Decoction on endotoxin levels of patients with cirrhosis endotoxemia due to hepatitis B. METHODS 58 patients with cirrhosis endotoxemia due to hepatitis B of toxic heat and blood stasis syndrome were randomly divided into treatment group(30 cases) and control group(28 cases). Patients in the control group were treated with hepatoprotective medication therapy, including reduced glutathione injection, Kuhuang injection and diammonium glycyrrhizinate and sodium chloride injection. Based on this, patients in the treatment group were further given Qingying Decoction. The endotoxin levels in the serum of these 58 patients were tested using the dynamic turbidity method, together with serum alanine aminotransferase(ALT), total bilirubin(TBiL) and prothrombin time(PT). RESULTS The treatment group was superior to the control group in decreasing clinical symptom integral(P<0.01) and even more superior in reducing endotoxin levels(P<0.05). CONCULSION Qingying Decoction has a notable curative effect on cirrhosis endotoxemia due to hepatitis B.
Influences of Qinzhu Liangxue Decoction on Serum IL-17, IL-23 and Related Transcription Factors of Skin Lesion in Patients with Psoriasis of Blood-heat Syndrome

Abstract:
OBJECTIVE To provide foundation for traditional Chinese medicine in treating psoriasis via studying the intervention effect of Qinzhu Liangxue Decoction on Th17 cells, STAT3 and RORγt of psoriasis patients. METHODS 60 cases of psoriasis vulgaris(of blood-heat syndrome) were selected for clinical research and randomly divided into treatment group(30 cases) and control group(30 cases), with the normal group being 10 cases. Patients in the treatment group and control group was prescribed with oral Qinzhu Liangxue Decoction and acitretin respectively, with the treatment lasting for 1 month. Clinical efficacy was observed based on psoriasis area and severity index(PASI). ELISA was adopted to detect, compare and analyze changes of Th17 cells. Expression rules of STAT3 and RORγt in the lesion tissue of psoriasis were detected by immunohistochemistry technology. RESULTS PASI scores of both groups after treatment were lower than those before treatment, and the difference was statistically significant(P<0.05). Serum Th17 cytokines(IL17, IL23) of the two groups before treatment were higher than the normal group(P<0.05), and both IL17 and IL23 of these two groups went down after treatment(P<0.05). Before treatment, the expression of STAT3 and RORγt via immunohistochemical detection in the lesion area of both groups was higher than that of normal group(P<0.05), while it became lower after treatment(P<0.05). CONCULSION Qinzhu Liangxue Decoction plays an effective role in treating psoriasis by improving Th17 cells and regulating transcription factors STAT3 and RORγt.
Influences of Reinforcing Qi and Nourishing Yin Method Combined with Gemcitabine on Quality of Life of Senile Patients with Advanced Non-small Cell Lung Cancer

Abstract:
OBJECTIVE To observe the influences of reinforcing qi and nourishing yin method combined with gemcitabine on immunologic function and quality of life in senile patients with advanced non-small cell lung cancer(NSCLC). METHODS 60 senile patients with advanced NSCLC were randomly divided into treatment group(31 cases treated with Yiqi Yangyin Kangliu Formula plus gemcitabine) and control group(29 cases given gemcitabine only). Such changes as quality of life, clinical symptoms, adverse reaction of chemotherapy and immunologic function of both groups during the observation period were recorded. RESULTS After treatment, body function scores, role function scores as well as the overall quality of life scores of patients in the treatment group were much higher than those in the control group(P<0.01). And such symptom scores as dyspnea, insomnia, loss of appetite in the treatment group were much lower than those in the control group(P<0.05~0.01). Symptoms like cough, shortness of breath, dry mouth and throat were reduced obviously, which were superior to control group(P<0.01). Compared to the control group, there were partial improvements of serum immune function in the treatment group(P<0.01); And adverse reaction of chemotherapy in the treatment group was lower than that in the control group(P<0.05). CONCULSION Reinforcing qi and nourishing yin method combined with gemcitabine is effective in treating senile patients with advanced NSCLC by improving clinical symptoms and quality of life and reducing side effects caused by treatment.
Effects of Sijunzi Decoction on Immunologic Function and Quality of Life in Patients with Rectal Carcinoma After Surgery

Abstract:
OBJECTIVE To explore the effects of Sijunzi Decoction on immunologic function and quality of life in patients with rectal carcinoma after surgery. METHODS 130 cases with rectal carcinoma having undergone surgery were randomly divided into study group(n=65) and control group(n=65). Patients in the control group were treated with FOLFOX7 chemotherapy regimen, while those in the study group were further given Sijunzi Decoction, in addition to the FOLFOX7 chemotherapy regimen, with the treatment lasting for 4 courses. Then changes of KPS scores, Chinese medical syndrome scores and T-lymphocyte subsets were evaluated in both groups. RESULTS After treatment, KPS scores in the study group were increased evidently but decreased notably in the control group, and there were statistical significances between two groups in changes of quality of life(P<0.05). Compared to the study group, the decreased levels of CD3+,CD4+, CD8+ and CD4+/CD8+ proportion in the control group after treatment were more obvious(P<0.05~0.01). When compared to the control group, the symptom scores like anorexia, spiritlessness, abdominal distension, loose stool as well as the total scores went down more significantly in the study group(P<0.05~0.01). Pearson correlation analysis showed that KPS scores were positively related to CD4+/CD8+ proportion(r=0.486, P<0.05), while TCM syndrome scores were negatively related to CD4+/CD8+ proportion(r=-0.375, P<0.05). CONCULSION The adjuvant chemotherapy of Sijunzi Decoction plus FOLFOX7 regimen is effective in treating postoperative rectal cancer by significantly improving the immunologic function, quality of life as well as the clinical symptoms.
The Effect of Astragaloside-Ⅳ on Oxygen Stress and EMT of HMrSV5 Induced by High-glucose Peritoneal Dialysate

Abstract:
OBJECTIVE To observe the effect of Astragaloside-Ⅳ (AS-Ⅳ) on oxygen stress and EMT of peritoneal mesothelial cells (PMCs) HMrSV5, explore the function of ROS in EMT and regulatory mechanism of AS-Ⅳ. METHODS We used peritoneal dialysis solutions (PDS) with high glucose concentration to establish oxygen stress cell model of HMrSV5. Morphologic alterations of cells were observed and MTT assay was used to detect cell viability. DHE fluorescent probes were loaded to label secreted ROS. EMT-associated proteins were expressed by western blotting analysis. RESULTS ①40 or 50μg/mL AS-Ⅳ increased the viability of HMrSV5 (P<0.05). Cell viability was supressed due to the longstanding effect of PDS and high glucose concentration, notably in intervention of PDS containing 4.25% glucose for 48h. Phenotypic changes of HMrSV5 characterized by cell elongation and losing cobblestone like feature. ②40μg/mL AS-Ⅳ could suppress morphologic changes of cells, decrease ROS expression in model group (P<0.05), upregulate the protein expression of E-cadherin and ZO-1 while downregulate α-SMA. Furthermore, the phosphorylation of Smad2/3 protein was inhibited. ③ 5mmol/L NAC had the similar effect on EMT of PMCs. CONCLUSION PDS with high glucose concentration can suppress cell viability and induce EMT of PMCs by upregulating ROS expression and activation of Smads pathway. AS-Ⅳ can reduce ROS generation, suppress the phosphorylation of Smad2/3 and thus, inhibite the EMT of PMCs.
Study on Function of JAK2/STAT3 Pathway in Promoting Angiogenesis with Angelica Microcapsules for Replenishing Blood

Abstract:
OBJECTIVE To investigate the function of JAK2/STAT3 pathway in promoting angiogenesis with Angelica microcapsules for replenishing blood (Ang-Mc). METHODS Establish the human umbilical vein endothelial cell (HUVEC) model, and then cell counting kit (CCK-8) and flow cytometry were used to test cell viability and cell apoptosis. Western blot was used to detect expression of p-STAT3 and VEGF. RT-PCR was used to detect expression of vascular endothelial growth factor (VEGF) and miRNA-21. RESULTS When Ang-Mc with a concentration of 20μg/mL acted on HUVEC for 24h, it could effectively promote the HUVEC proliferation by up-regulating the expression of p-STAT3, VEGF and miRNA-21. AG490 can effectively inhibit Ang-Mc promoting HUVEC proliferation effect and reduce the expression of p-STAT3, VEGF and miRNA-21. CONCLUSION The effect of Ang-Mc in promoting angiogenesis is related to JAK2/STAT3 signaling pathways.
Mechanism of thermoTRP Participating in Cold Hyperalgesia of Rat with Experimental Knee Osteoarthritis

Abstract:
OBJECTIVE To observe both TRPA1 and TRPM8 expressions in synovial tissues of rats with cold hyperalgesia induced by experimental knee osteoarthritis(KOA). METHODS 45 Sprague Dawley male rats, weight ranging from 440g to 470g were randomly individed into the model group, modelantagonist and normal group. Paw withdrawal latency (PWL) of five rats from each group was determined one week before modeling, 2, 4, 6 and 8 weeks after modeling, respectively. 8 weeks after modeling, the rats were killed and synovial and cartilage tissues from diseased knee were collected, to observe pathological morphology at cartilage tissues and to determine the protein and gene expressions of TRPA1 and TRPM8 at synovial tissues. RESULTS KOA rats showed obvious cold hyperalgesia from two weeks after modeling to the latest followup, 8 weeks after modeling. The abnormally low level of PWL could be increased by TRPA1 and TRPM8 ion channel blocker. CONCLUSION Upregulating expressions of TRPA1 and TRPM8 participate in the occurrence mechanism of KOA cold hyperalgesia.
Impact of "Zao-ji-sui-yuan" Combined with Glycyrrhizae on Rat Intestinal P-glycoprotein Transport Function in Vitro

Abstract:
OBJECTIVE The mechanism of Glycyrrhizae's incompatibility with "zao-ji-sui-yuan"(Sargassum, Euphorbia pekinensis, Euphorbia kansui and Daphne genkwa) was explored by assessing intestinal P-glycoprotein (P-gp) activity in vitro. METHODS Ussing-chamber technique was used to determine intestinal secretory function. Rhodamine123 (R123) was used as an indicator of P-gp activity. RESULTS Glycyrrhizae could significantly promote the secretion of R123. In the present of Sargassum, Euphorbia pekinensis and Daphne genkwa, the promotion effect of Glycyrrhizae was significantly suppressed. CONCLUSION P-gp activity can be significantly induced by Glycyrrhizae. The promotion effect of P-gp activity induced by Glycyrrhizae was antagonized by Sargassum, Euphorbia pekinensis and Daphne genkwa, which might lead to the reduction of toxic components pumping out and then aggravated intestinal tissue injury. These results indicated that when Glycyrrhizae combined with some toxic traditional Chinese medicine, change of the intestinal P-gp activity might cause the "contrary effects".
Establishment and Staging Studies of Human Ovarian Cancer HO-8910 Orthotopic Xenograft Animal Models

Abstract:
OBJECTIVE To establish an ovarian cancer xenograft model by fluorescence and study of the cancer staging. METHODS The human ovarian cancer HO-8910 cells were cultured for 10 generations in the cell culture dish, and the cells that could stably express fluorescence were selected and injected into nude mice. When the tumor grew to a mass volume of 1mm3, it was implanted into the ovarian mucous membrane of nude mice through micro-surgery. The tumor growth and metastasis were observed by in vivo imaging technique, and the tumor tissue and surrounding tissues were observed through pathology detection methods. RESULTS In vivo imaging technology successfully traced the development and metastasis of tumor. Pathological examination showed tumor cell metastasis trend. Both of them combining with TCM clinical ovarian cancer staging determined the ovarian cancer staging. CONCLUSION The experiment successfully establishes ovarian cancer orthotopic transplantation model and represents the evolution of clinical oncology.The stages will provide some experimental basis for the study of tumor and the evaluation criterion of antitumor drug effect.
Protective effects of cyclovirobuxine D on myocardial ischemic in rats by Micro-PET

Abstract:
OBJECTIVE To evaluate the protective effects of Cyclovirobuxine D on myocardial ischemic rats by the method of Micro-PET scanning technology. METHODS All male rats were randomly divided into sham group, model group, digoxin group and Cyclovirobuxine D group. The myocardial ischemic models were established in all groups except sham group. The imaging agent 18FFDG was injected intravenously into tail in rats on the first day of the model and 7th day during medication. The Standard Uptake Value(SUV) of the heart was measured by the method of Micro-PET scanning technology. The changes of myocardial infarction rate were evaluated by Volume of Interest (VOI) sketch. RESULTS The levels of SUV of the heart were improved and the myocardial infarction rates were reduced in digoxin group and Cyclovirobuxine D group(P<0.05~0.01), compared with the model group. CONCLUSION Cyclovirobuxine D has protective effects on myocardial ischemic rats, which can be concluded by the Micro-PET imaging technology in living conditions of rats.
Mechanism of Wenwei Tiaochang Granule on Rats with Diarrhea-Predominant Irritable Bowel Syndrome

Abstract:
OBJECTIVE To explore the effects and mechanisms of Wenwei Tiaochang Granule on diarrhea-predominant irritable bowel syndrome rats. METHODS 60 SD rats, were randomly divided into control group, model group, positive drug group, and low, middle and high dose groups of Wenwei Tiaochang Granule. In order to make the rats get diarrhea-predominant irritable bowel syndrome, the rats were administrated with senna, bound limbs and clipped tails for 14d. After 14d of administration with positive drug and Wenwei Tiaochang Granule, the blood, jejunum, colon, thymus and spleen were collected. Motilin assay kit was used to detect motilin (MTL) content in plasma and jejunum. Use the weight of thymus and spleen to calculate their organ index. Calcitonin gene-related peptide(CGRP) and substance P (SP) mRNA level and protein expression level was detected by RT-PCR and Western-blot in colonic tissues. RESULTS Compared with the control group, the growth rate and food intake increased slower in model group, at the same time, the thymus index, spleen index and MTL content in plasma and jejunum decreased significantly, while the CGRP and SP mRNA and protein expression level of colon increased significantly (P<0.05~0.01). Compared with the model group ,the middle and high dose groups of Wenwei Tiaochang Granule could significantly increase the thymus index, spleen index and MTL content in plasma and jejunum, and decrease CGRP and SP mRNA and protein expression level of colon(P<0.05~0.01). CONCLUSION By regulating the content of MTL and the expression level of CGRP and SP mRNA and protein, Wenwei Tiaochang Granule can be used to treat diarrhea-predominant irritable bowel syndrome.
Secondary Metabolites from the Endophyte Fungus Aspergillus sp.Tpxq

Abstract:
OBJECTIVE To study the secondary metabolites of an endophytic fungus Aspergillus sp. TPXq isolated from Saussurea medusa. METHODS The strain was cultured in a large-scale by using the shaking incubator. The secondary metabolites were isolated by chromatography methods such as silica gel, Sephadex LH-20 columns, ODS flash column and HPLC, etc. The structures of these compounds were identified by their physico-chemical constants and the NMR data. RESULTS Eight Cyclodipeptides and one alkaloidwere seperated from secondary metabolites of an endophytic fungus Aspergillus sp. TPXq isolated were from Saussurea medusa., and their stuctures were identified as 3-isobutylpyrrolopiperazine-2,5-dione (Ⅰ),3-isopropyl-pyrrolopiperazine-2,5-dione (Ⅱ), 3-seco-butyl-pyrrolopiperazine-2,5-dione (Ⅲ), 3-benzyl-pyrrolopiperazine-2,5-dione (Ⅳ), 3-benzyl-6-(p-hydroxy benzyl)piperazine-2,5-dione (Ⅴ), 3,6-dimethylpiperazine-2,5-dione (Ⅵ), 3-isobutyl-6-isopropylpiperazine-2,5-dione (Ⅶ), 3-isobutyl-6-benzylpiperazine-2,5-dione (Ⅷ), Chaetominine (Ⅸ), by analysis of physiochemical and NMR data. CONCLUSION Among them, compounds Ⅳ~Ⅷ were isolated from Alterraria.sp for the first time. Compounds Ⅸ showed significant cytotoxic activity against A549 and MCF-7 tumor cells, the IC50 values are 0.18μg/mL and 0.89μg/mL, respectively. However, compoundsⅠ~Ⅷ exhibited very weak cytotoxicity (>50μg/mL) against these cell lines.
The Application of IP-RPHPLC in Determination of ATP, ADP and AMP in Liver of the Rats Treated by Electric Acupuncture

Abstract:
OBJECTIVE To establish an ion-pair reversed-phase high performance liquid chromatography(HPLC) method and determine ATP, ADP and AMP in liver of the rats treated by electric acupuncture. METHODS Separation of ATP, ADP and AMP in the liver tissue samples of rats was performed on a Megres-C18 water resistant column (5μm, 250mm×4.6mm) by Shimadzu SIL-20AT. A mobile phase was 0.18mol/L phosphate dihydrogen containing 5% methanol (pH=6.25). The detection wavelength was set at 254nm and the flow rate was 0.8mL/min. The injection volume was 20μL and the column temperature was set at 25℃. RESULTS The linear ranges of ATP, ADP and AMP were 1.1~212.2μg/mL(r=0.9999), 0.9~180μg/mL(r=0.9999) and 0.9~181.8μg/mL (r=0.9997). The average recovery rates of ATP, ADP and AMP were 96.4%, 98.3% and 100.8%; and RSD were 2.50%, 2.88% and 4.14%, respectively. The RSD of intra-day and inter-day were 0.06%~0.40% and 0.06%~0.69%. The detection limits for ATP, ADP and AMP were 0.11,0.09μg/mL and 0.045μg/mL, respectively. CONCLUSION The established method for the determination of ATP, ADP and AMP in the liver of rats is simple, quick, accurate and reliable, which will lay a foundation for the determination of small molecular bio-active substances in the animal tissues treated by acupuncture intervention.
Study on the Relationship Between the Deformability of Transfersome Containing Coumarin-6 and Transdermal Permeation Properties in Vitro

Abstract:
OBJECTIVE To study the relationship between the indexs of the deformability of the different prescription of transfersomes containing coumarin-6 and transdermal permeation properties in vitro. METHODS Four kinds of different prescription of transfersomes containing coumarin-6 were prepared by using film dispersion method. The formulation was evaluated through vesicle size, vesicle size distribution, drug entrapment efficiency, deformability and rat skin permeation amount in vitro. The deformability of vesicles was measured with extrusion method and evaluated by such three different indexes as the deformation index D,the corrected deformation index Dcor and the index RD. RESULTS The evaluation index D and Dcor shows that the order of the deformability of four different prescription of transfersomes containing coumarin-6 is sodium deoxycholate(TF-SDC)>sodium cholate(TF-SC)>the blank (Liposome)>span 80(TF-SP80) and the evaluation index RD shows that the deformability order is TF-SP80>TF-SDC>TF-SC>Liposome; the order of cumulative penetration amount of coumarin-6 in 12h is TF-SDC>TF-SC>Liposome>TF-SP80. CONCLUSION The evaluation index D and Dcor have the best correlation with the cumulative penetration amount of coumarin-6 in vitro among 3 kinds of deformability index.
Study on Limonin Distribution in Rat Intestine by LC-MS/MS Method

Abstract:
OBJECTIVE Integral animal experiment was employed to investigate the limonin distribution in different parts of rats’ intestine. METHODS The concentration of limonin in different parts of the intestinal was detected by LC-MS/MS method. After the rats were given the limonin 80mg/kg ig, the drug concentrations in the duodenum, small intestine and rectum were detected at 2,3,4,6 and 8h. RESULTS Limonin was highly distributed in different parts of the intestinal tissue. Most of intestinal tissue reached peak concentration at 3~4h after administration. The drug concentration in the duodenum and rectum was the highest, with duodenum being the second. CONCLUSION Limonin has a wide distribution and high concentration in rat intestine, and it plays a direct role in the treatment of hemorrhoids in intestine.
Optimization of Extraction Technique of Seed Oil from Annona squamosa

Abstract:
OBJECTIVE To determine the optimal parameters in extraction of seed oil from Annona squamosa. METHODS The extraction efficiency of four extraction methods, including percolation extraction, soxhlet extraction, ultrasonic extraction and SFE-CO2 extraction were compared. The optimal parameters of the best extraction method were determined, and the chemical composition of the oils was analyzed by GC-MS. RESULTS The yields of the oils extracted by the soxhlet extraction method were highest (22.28±0.21%), and the optimal parameters were obtained as follows: Petroleum ether were used as extraction solvent, the extraction temperature was 80℃, and the extraction time was 90 min. GC-MS analysis results showed that the main chemical composition was palmitic acid (9.92%), linoleic acid (20.49%), oleic acid (56.50%) and stearic acid (9.14%). CONCLUSION The optimum extraction method was stable. Annona squamosa seeds are rich in unsaturated fatty acids and have a certain economic value.
Clinical Study of Yangyin Runmu Pill for Treating Dry Eye Disease

Abstract:
OBJECTIVE To study the clinical effects of Yangyin Runmu Pill in the treatment of dry eye disease. METHODS 120 cases with dry eye disease were randomly divided into two groups with 60 cases in each group. Patients in the treatment group were treated with oral Yangyin Runmu Pill and those in the control group were given Tears Naturale Forte Eye Drops. And the curative effects of two groups were to be compared after 20 days of treatment. RESULTS The total effective rates of the treatment group and the control group were 86.7% and 63.3% respectively, with a statistical significance(P<0.05). In the treatment group, Schirmer test(SIT) showed an obvious increase of lacrimal secretion and also the tear break-up time(BUT) was extended obviously after treatment, which displayed a significant difference from those before the treatment as well as those in the control group of the same period(P<0.01). The ocular surface staining of the treatment group after treatment was obviously better than that of the control group (P<0.01). CONCULSION Yangyin Runmu Pill can improve ocular surface symptoms effectively and has exact curative effects for dry eye disease, which makes it worthy of clinical application and promotion.
Clinical Observation on Treating Stiff Neck by Acupuncture Based on Meridians and Exercise Therapy

Abstract:
OBJECTIVE To observe the clinical effects of acupuncture based on meridians and exercise therapy in treating stiff neck. METHODS 62 patients with stiff neck were randomly divided into treatment group and control group according to the doctor order with dual number, with 31 cases in each group. Patients in the treatment group were treated with acupuncture based on meridians and exercise therapy, while those in the control group were given acupuncture therapy. Scores of VAS and active degrees of cervical vertebrate were used to evaluate effects. RESULTS After treatment, there were statistical significances in both VAS scores(P<0.01) and active degrees of cervical vertebrate(P<0.01) of two groups, with the treatment group being superior to the control group(P<0.05). CONCLUSION Effects of acupuncture based on meridians and exercise therapy on stiff neck are obvious.
