Abstract:
This paper introduces clinical experience of Prof. Xuan Guowei in treating acne caused by hyperactivity of fire due to yin deficiency by applying method of nourishing yin and clearing away heat. Prof. Xuan Guowei pointed out that the cause of the acne was the kidney yin deficiency, which resulted in the imbalance of yin and yang of kidney and the hyperactivity of ministerial fire, thus causing the blood heat in lung and stomach. The treating principal is nourishing yin to discharge fire and clearing lung and removing toxicity. Xiaocuo decoction made by the modification of Erzhi pills together with Zhibai Dihuang pills can be used to treat acne.

Abstract:
Since The Yellow Emperor's Inner Classic put forward the concept of retrograde treatment, physicians from different period of history had studied it from different perspectives. According to the feature of Inner Classic, the concept of retrograde treatment must be concise, which means that it is the opposite treating method based on reverse thinking. Wang Mengying was the famous doctor of epidemic febrile disease of later Qing Dynasty. When treating diseases, he would find the roots and it was quite useful in the treatment.

Abstract:
OBJECTIVE To investigate the effects of Whenshen Zhixie Fang administrated through bellybutton and the influence on 5-HT and NO of spleen and kidney yang deficiency type IBS-D patients. METHODS By using computer, parallel group and non-blinded trials, patients were randomly divided into two groups. 58 cases in the treatment group were given Wenshen Zhixie Fang administrated through bellybutton together with orally taken Montmorillonite powder. The control group was given orally taken Montmorillonite powder only. The treating effects were assessed after ten days. RESULTS Total effective rate of the treatment group was 82.76%, which was better than that of the control group (66.10%)(P<0.05). Abdominal pain and discomfort were significantly relieved in the treatment group, and the shape of the stool and frequency were also changed for better (P<0.01), which were better than those of the control group except the stool. And 5-HT was significantly decreased and NO was increased, which were better than those of the control group(P<0.01).CONCLUSION Wenshen Zhixie Fang administered through Bellybutton can improve the clinical symptoms of IBS-D and its mechanism may be related to the regulation of 5-HT and NO levels and reduction of intestinal sensitivity.
Evaluation on Clinical Therapeutic Effects of Modified Zhenwu Decoction in Treating Ⅳ Stage Diabetic Nephropathy Due to Yang Deficiency of Spleen-kidney

Abstract:
OBJECTIVE To explore the clinical therapeutic effects of Jiawei Zhenwu Decoction in treating IV stage diabetic nephropathy(DN) due to yang deficiency of spleen-kidney. METHODS 130 cases with DN at stage Ⅳcollected from January, 2014 to June, 2015 were randomly divided into study group and control group, with 65 cases in each group. Conventional therapy were applied to both groups and based on this, patients in the study group were further given Modified Zhenwu Decoction. After the treatment for one month, changes of TCM syndrome scores, fasting blood-glucose(FPG), urine protein excretion, serum creatinine(Scr), creatinine clearance rate(Ccr), systolic blood pressure(SBP) and diastolic blood pressure(DBP) before and after treatment were observed and comparision were made between these two groups to evaluate the clinical efficacy and safety. RESULTS Compared to the control group, there were more evident declines of TCM syndrome scores, FPG, urine protein excretion, Scr, Ccr, SBP and DBP in the study group after treatment, with statistical differences(P<0.05). The total effective rate of study group was 90.77%, which was much higher than that of 67.69% in the control group, and there were statistical differences between two groups(P<0.05). No serious side effects were noticed during the treatment in both groups. CONCULSION Conventional therapy plus Modified Zhenwu Decoction can evidently improve clinical symptoms and kidney function of patients with IV stage DN due to yang deficiency of spleen-kidney, with no side effects.
Longdan Xiegan Decoction in the Treatment of Embolism Syndrome after Interventional Therapy on Primary Hepatocellular Carcinoma

Abstract:
OBJECTIVE To observe the effect of Longdan Xiegan decoction in treating embolism syndrome after interventional therapy on primary hepatocellular carcinoma. METHODS Sixty patients with primary cancer after interventional therapy were randomly divided into two groups: the treatment group(30 cases) and the control group(30 cases). The control group was treated by western medicine and the treatment group, besides taking western medicine, took oral-taken Longdan Xiegan decoction as well for six weeks. RESULTS The effective rates of both groups showed no significant difference(P>0.05). After one week of treatment, the severity degree of the treatment group was much lower than that of the control group(P<0.05). After two weeks' treatment, the TCM score of the treatment group was much lower than that of the control group(P<0.05). The treatment group showed significant improvement in the TCM syndromes(P<0.05). The life quality of the treatment group was much better than that of the control group after two weeks' treatment(P<0.05). After two weeks' treatment, the treatment group showed significant decrease in the liver function(P<0.05). CONCLUSION The combination of Longdan Xiegan decoction and western medicine are more effective in treating embolism syndrome after interventional therapy on primary hepatocellular carcinoma.
Clinical Research on Transcutaneous Electrical Stimulation at Yifeng Acupoint(TE17) Combined with Stellate Ganglion Block in Treating Hunt Syndrome at Acute Stage

Abstract:
OBJECTIVE To observe the clinical therapeutic effects of transcutaneous electrical stimulation at Yifeng acupoint(TE17) combined with stellate ganglion block in treating Hunt syndrome at acute stage and their influence on levels of stress hormones. METHODS 56 cases diagnosed with Hunt syndrome at acute stage were divided into observation group and control group based on consultation time, with 28 cases in each group. Patients in the control group were given conventional therapy(oral famciclovir granules and mecobalamin capsules), while those in the observation group were further given transcutaneous electrical stimulation of Yifeng acupoint(TE17) and stellate ganglion block, apart from the conventional therapy. After two courses of treatment, pain degree, regression time of herpes, recovery time of facial paralysis, adrenaline and noradrenaline content were observed and comparison were made before and after treatment.RESULTS Pain degree(vas pain scores), regression time of herpes, recovery time of facial paralysis in the observation group after treatment were lower or less than those in the control group, with statistical differences(P<0.05). There were statistical differences before and after treatment in adrenaline and noradrenaline content of both groups(P<0.05). And statistical differences could be found between the two groups when compared with each other(P<0.05). CONCLUSION Transcutaneous electrical stimulation of Yifeng acupoint(TE17) combined with stellate ganglion block possesses certain therapeutic effects in treating Hunt syndrome at acute stage and is able to reduce stress responses so as to avoid other complications.

Abstract:
OBJECTIVE To observe changes of pressing pain along governor vessel on the back(below the spinous process from the 3rd cervical vertebra to the 9th cervical vertebra) in patients with gastro-esophageal reflux disease(GRED) after acupuncture treatment as well as its clinical meaning. METHODS 60 cases of GRED were randomly divided into acupuncture group and medication group, with 30 cases in each group. In the acupuncture group, such acupoints as Shenzhu(GV12), Shendao(GV11), Lingtai(GV10), Zhiyang(GV9), Jinsuo(GV8) and non-acupoints(T4, T8) were needled once every other day, three times a week. While those in the medication group were given Omeprazole(20mg/time) twice a day, with the treatment course lasting for eight weeks. Reflux disease questionnaires (RDQ) were answered in both groups before and after the treatment, then pain degree and pain threshold and pressing pain along governor vessel from T3~T9 were assessed. RESULTS The effect rate was 70% in acupuncture group and 43.3% in medication group with significant differences between the two groups (P<0.05). After treatment, the relief of pressing pain below the spinous processes from T3 to T9 in acupuncture group was better than that in medication group(P<0.05), and the increasing of pain threshold from T5 to T7 in acupuncture group was much higher than that in medication group(P<0.05). CONCLUSION Acupuncture stimulation along governor vessel on the back is more effective than medication, and the pain threshold after acupuncture was significantly higher than medication. Therefore, the changes of pressing pain along governor vessel can be considered as an index to estimate the effect of treating GRED by acupuncture.

Abstract:
OBJECTIVE To observe the effect of Sanhuang decoction in the body mass and metabolic index of patients with obesity. METHODS 64 Patients conformed to western medicine diagnosis of obesity and phlegm, dampness and heat junction were randomly divided into Sanhuang decotion group and lifestyle intervention group. After three months of treatment,comparative analysis of clinical symptoms,body mass index, waist, glucose and lipid metabolism factors and inflammatory cytokine were carried out. RESULTS Sanhuang decoction group showed better improvement in lowering body mass, waist, TG levels than the control group(P<0.05). CONCLUSION Sanhuang decoction is better in treating obesity.

Abstract:
OBJECTIVE To study the effects of Qingluo Tongbi Compound(QLT) on pain behavior and COX-2 mRNA expression in dorsal root ganglion(DRG) and blood PGE2 concentration, and to explore the mechanisms of pain in rheumatoid arthritis. METHODS The mice were randomly divided into control group, celecoxib 30mg/kg,QLT 4.35g/kg, QLT 8.70g/kg and QLT 17.4g/kg groups. The mice pain threshold change were measured by hot plate method, and body torsion times were tested by acetic acid twisting method. The collagen-induced arthritis(CIA) was induced by collagenⅡ in DBA/1 mice. The mice were randomly divided into control group, CIA group, celecoxib 30mg/kg group, QLT 8.70g/kg. The paws swelling, mechanical withdrawal threshold(MWT) and thermal withdrawal latency(TWL) were recorded. After treated with corresponding drugs by intragastric administration for four weeks, COX-2 mRNA in DRG and PGE2 in mice were quantified. RESULTS QLT reduced pain threshold and body torsion times in pain model mice(P<0.01); compared with the CIA group, the QLT group decreased the paws swelling(P<0.05), up-regulated MWT(P<0.01) and PWL(P<0.05), down-regulated the expression level of COX-2 mRNA in DRG and PGE2 concentrations in blood. CONCLUSION QLT shows certain analgesic action, which might be related to the inhibitory effect of COX-2 mRNA expression in DRG and PGE2 in blood.
Effect of Acorus and Curcuma Decoction on Cortical Hormone in Mice Hippocampus NF-L and SYP Protein

Abstract:
OBJECTIVE To investigate the reverse effect of Acorus and Curcuma Decoction on despair like behavior in mice induced by hypercortisolism. METHODS 60 C57BL/6N mice were randomly divided into 6 groups: normal control, model control, Fluoxetine hydrochloride group, Acorus and Curcuma Decoction groups of high dose, middle dose and low dose. The hypercortisolism mice models were developed by subcutaneous corticosterone injection. Four weeks later, the changes of mice body, spleen and thymus mice weight were detected. After the forced swimming test and the tail suspension test finished, the mice were sacrificed by decapitation and the hippocampal tissues were rapidly collected for Western blot analysis of neurofilament light chain protein(NF-L) and synaptic vesicle protein(SYP). RESULTS Compared with the normal control group, the body weight was lower than model control group(P<0.05), the immobility time was significantly prolonged in the forced swimming test and the tail suspension test (P<0.01), spleen and thymus weight significantly decreased (P<0.05), the expression of NF-L and SYP, were down-regulated in model control group(P<0.01). However, Acorus and Curcuma Decoction group, significantly shortened immobility time in behavioral science(P<0.01), and up-regulated the expression of NF-L and SYP (P<0.01). CONCLUSION Acorus and Curcuma Decoction can protect neurons and antagonized the despair like behavior induced by hypercortisolism.
Influences of Anzi Heji on JAK/STAT3 Signaling Pathway on the Maternal-Fetal Interface of Mice Suffering ACA Positive Abortion

Abstract:
OBJECTIVE It is to observe the influences of Anzi Heji on the JAK/STAT3 signaling pathways on the maternal-fetal interface of anticardiolipin antibody (ACA) positive mice. METHODS Establish ACA positive mice model with humanβ2-GPⅠ as derivant, and administer normal saline to the blank group and humanβ2-GPⅠ to the model group; administer aspirin and high and low dose Anzi Heji respectively to each therapy group, kill the mice at the 15th day of their pregnancy, calculate the embryo resorption rate, and test the ACA of peripheral blood by means of ELISA; test the JAK, GP130 and p-STAT3 of placenta and decidual tissue with immunohistochemical method, and test STAT3 and p-STAT3 with western blot method. RESULTS The embryo resorption rate of the mice of low- and high-dose Anzi Heji groups and aspirin group was lowered obviously, so was the titer of ACA in the serum of mice (P<0.05), the expression of JAK, GP130 and p-STAT3 on the maternal-fetal interface was raised, and wherein, the expression of JAK, p-STAT3 of placenta had significant difference from that of model group (P<0.05), and the expression of GP130 and p-STAT3 of decidual tissue had significant difference from that of the model group (P<0.05). CONCLUSION The pathomechanism for ACA to induce pregnancy loss is related to the JAK/ STAT3 signaling pathways on the maternal-fetal interface, and the mechanism for Anzi Heji to produce therapeutic effect is possibly to promote the activation of STAT3 of placenta and decidual tissue by enhancing the expression of JAK of placenta and strengthening the expression of GP130 of decidual tissue.
Effects of Isoliquiritigenin on Induction of Apoptosis and Expression of ERS Related Genes in NCI-H157 Cells

Abstract:
OBJECTIVE To study the effect of isoliquiritigenin on cell proliferation, apoptosis induction, mRNA expression of endoplasmic reticulum stress related gene ATF4, DDIT3 and TNFRSF10B in NCI-H157 human lung cancer cell line. METHODS NCI-H157 cells were cultured and treated with various concentrations isoliquiritigenin. The cell proliferation was analyzed by MTT and the occurrence of cell apoptosis was examined by flow cytometry. ATF4, DDIT3 and TNFRSF10B gene mRNA expression were detected by real-time fluorescence quantitative polymerase chain reaction. RESULTS Isoliquiritigenin significantly inhibited NCI-H157 cell proliferation in a dose and time dependent manner. The results of flow cytometry revealed that isoliquiritigenin could induce apoptosis in NCI-H157 cells. Real-time quantitative RT-PCR showed that isoliquiritigenin up-regulated mRNA expression of ATF4, DDIT3 and TNFRSF10B(P<0.05~0.01). CONCLUSION Isoliquiritigenin can inhibit NCI-H157 cell proliferation, induce cell apoptosis by activating the endoplasmic reticulum stress response.
Intervention Effect of Taishan Panshi San on Early Pregnancy Loss in PCOS Rats with Ovulation Induction

Abstract:
OBJECTIVE To investigate the possible mechanism of early pregnancy loss in PCOS rats with ovulation induction and the intervention effect of Taishan Panshi San on pregnancy loss. METHODS To select normal pregnant rats mated with male rats as group A. The PCOS rats model by dehydroepiandrosterone(DHEA) was stimulated ovulation by clomiphene(CC),then mated with male rats to establish the rat model of pregnancy.They were randomly divided into two groups:treated by normal saline (group B) and by Taishan Panshi San (group C). According to the pregnancy third days, fourth days, fifth days, group B was divided into group B3, group B4, group B5. To detect the mRNA and protein expression of endometrium HOXA10, integrinαvβ3, EMX2, LIF by RT-PCR and Western blot in each group. RESULTS In group B of PCOS Pregnant rats with ovulation induction:The expression of mRNA HOXA10 in Group B4 was higher than group B3 and group B5(P<0.05~0.01),the expression of mRNA integrinαvβ3 was significantly higher than group B3 and group B5(P<0.01),but the expression of mRNA EMX2 was lower than group B5(P<0.05);the expression of mRNA LIF was highest in group B3, higher than group B4(P<0.05)。In the planting window period:the mRNA expression of endometrium HOXA10, integrinαvβ3, LIF in Group B5 were significantly lower than Group A and Group C (P<0.05~0.01);the expression of endometrium EMX2 in Group A was significantly lower than Group B5 and Group C (P<0.01);the protein expression of endometrium HOXA10, integrinαvβ3, LIF in Group B5 were significantly lower than Group A and Group C ,but the protein expression of EMX2 higher than Group A and Group C (P<0.01). CONCLUSION PCOS rats with ovulation induction occurred early pregnancy loss may be related to the low expression of endometrial HOXA10,integrinαvβ3, LIF and the high expression of EMX2 in the implantation window phase. This may also be related to the abnomal expression of HOXA10, integrinαvβ3, LIF, EMX2 before the planting window.Taishan Panshi San improved the endometrial receptivity and reduced the early pregnancy loss,probably by raising the expressions of endometrium HOXA10, integrinαvβ3, LIF and decreasing the expressions of EMX2 in the implantation window phase.
The Effect of Yangxin2 Fang on the Myocardial ATP and MMP in Rats with Chronic Heart Failure

Abstract:
OBJECTIVE To study the effect of Yangxin2 Fang on the myocardial energy metabolism in rats with chronic heart failure(CHF), and to explore the mechanism of Yangxin2 Fang for the treatment of CHF. METHODS 45 male clean grade SD rats were divided into two groups, normal group(group A,n=6) and CHF group(n=39).The CHF group was injected hypodermic with ISO for 28 consecutive days. After the model was successfully established in CHF, the survival rats were randomly divided into three groups: model group (group B), Carvedilol group (group C)and Yangxin2 Fang group(group D).Group C and group D were received gastric gavage of Carvedilol and Yangxin2 Fang respectively. Group A and group B were received gastric gavage of distilled water. After 28 days of dosing, the levels of myocardial adenosine triphosphate (ATP)and mitochondrial membrane potential (MMP) were tested. RESULTS ①the change of ATP level: Compared with the group A, the level of myocardial ATP was decreased in group B(P<0.05); Compared with the group B and C, the level of myocardial ATP was increased in group D(P<0.01).Compared with group C, the level of myocardial ATP was increased in group D(P<0.01). ②the change of MMP levels: Compared with group A, the levels of MMP was decreased in group B(P<0.01) Compared with the group B, the levels of myocardial MMP were increased in group C and D.(P<0.01). CONCLUSION Yangxin2 Fang could increase the levels of myocardial ATP and MMP to improve myocardial energy metabolism, which might be one of the functionary mechanism of Yangxin2 Fang to treat CHF.
Study of the Effect and Mechanism of Dermatitis Lotion in the Treatment of Allergic Contact Dermatitis in Mice

Abstract:
OBJECTIVE To study the efficacy of dermatitis lotion in the treatment of allergic contact dermatitis in mice. METHODS The 2,4-dinitro chlorobenzene(DNCB) was applied on the dorsum of mice to induce the allergic contact dermatitis models. 60 mice were randomly divided into control, model(ACD), momestasone furoate, dermatitis lotion (0.4, 0.2, 0.1g/mL) groups, with 10 mice in each group. Mice were sensitized by DNCB and then treated topically with different drugs on lesions. The inflammation degree of skin lesion were observed, histopathological analysis was measured by HE strain and the levels of serum IFN-γ and IL-4 were detected by ELISA. RESULTS The dermatitis lotion of 0.4g/mL restrained the increase of thymus index, alleviated the inflammation degree of lesion on dorsum of mice and reduced the pathological score significantly, compared with the model group. In addition, the dermatitis lotion of 0.4g/mL adjusted the expression of serum IFN-γ and IL-4 to return to normal levels. CONCLUSION These results suggest that dermatitis lotion can be used for the treatment of allergic contact dermatitis (ACD) mainly through the regulation Th1/Th2 cytokines.
Pharmacodynamics of Changkang Granules in IBS-D Rats

Abstract:
OBJECTIVE To study the therapeutic effect of Changkang Granules in rat with irritable bowel syndrome with Diarrhea (IBS-D) and investigate its mechanism. METHODS The model for IBS-D was established by colorectal distention daily in the neonatal rats between postnatal 8 and 21 days. Sixty SD rats were divided into the normal control group,the model group,positive control group,high-dose,middle-dose,low-dose Changkang Granules group.The treatment lasted for 30 days. The threshold of abdominal withdrawal reflex(AWR) during rectal distension was evaluated. The calcitonin gene-related peptide(CGRP) in serum, vasoactive intestinal peptide(VIP) in colon homogenates were evaluated by radioimmunoassay.Immunohistochemical SABC method to observe expression level of SP of colonic specimens. RESULTS Compared with the normal control group,AWR score increased(P<0.01), The content of serum CGRP, VIP levels in colonic homogenate and expression level of SP in colon decreased markedly in the model group(P<0.01). After administration with Changkang Granules, the AWR scores of the IBS-D rats were reduced(P<0.01). The content of serum CGRP, colon VIP level and expression of SP in colon was increased significantly(P<0.05~0.01). CONCLUSION The therapeutic effect of Changkang Granules on IBS-D rat is obvious. The mechanism could be related to inhibition against the excessive decrease of serum CGRP, colon VIP and expression level of SP in colon in IBS-D rats.
Explore the Mechanism of Chinese Medicine Theory “Salt-hurt-blood” Based on High Salt-induced Kidney Injury in SHR Mice

Abstract:
OBJECTIVE To study the related basic research of high salt-induced kidney injury in SHR rats, and discuss the salt-hurt-blood connotation of traditional Chinese medicine. METHODS WKY rats were set as the normal group. SHR were randomly divided into 2 groups: normal group (SHR) and high-salt feed group (SHR+HS). SHR+HS group rats were given 4% high salt diet from the 10th week. Urine trace albumin and NAG in urine were detected every two weeks. On the 20th week, the hemorheology indexes and coagulation four indices were measured. The rat kidney was divided into two parts. One was fixed with formalin and detected with HE; the other part was used to detect ICAM-1, VCAM-1 and IL-6 expressions. RESULTS After feeding high-salt feed for 6 weeks, urine trace albumin and NAG increased significantly(P<0.05). On the 20th week, the whole blood viscosity and blood plasma viscosity in SHR+HS group were significantly(P<0.05) greater than SHR group. APTT and PT were significantly reduced and FIB significantly increased(P<0.05). Renal pathology index in SHR+HS group was higher than SHR group, and the kidney inflammation factors ICAM 1, VCAM 1 and IL-6 increased(P<0.05). CONCLUSION High salt can increase the whole blood viscosity, disorder APTT, PT and FIB, increase the expression of ICAM 1, VCAM 1 and IL-6, and strengthen the kidney injury of SHR rats, which might partly explain the scientific connotation of "salt-hurt-blood" and "salty flavor entering the kidney".
Lignans and Phenolic Compounds from Asparagus Gobicus

Abstract:
OBJECTIVE To study the chemical constituents of Asparagus Gobicus. METHODS Whole herb of Asparagus gobicus were extracted with 95% alcohol. Repeated silica gel column chromatography,Sephadex LH-20 chromatography, preparative HPLC and thin layer chromatography were applied for chemical constituents isolation and purification. Chemical structures of the compounds were elucidated by physical properties and spectra analysis such as NMR and ESI-MS. RESULTS Nine compounds were isolated from the extract of Asparagus gobicus. Six compounds were identified as lignans: (+)-pinoresinol(1), (-)-syringaresinol(2), (+)-syringaresinol-4'-O-β-D-glucopyranoside(3), (-)-episyringaresinol(4), (+)-nyasol(5), (+)-4'-O-methylnyasol(6). Three compounds were phenolic compounds: ferulic acid(7), isoferulic acid(8), caffeic acid(9). CONCLUSION All compounds were isolated for the first time from this plant,and compounds 1~4 and 8 were obtained from the genus Asparagus for the first time.
Comparison of Amino Acid Composition among Three Kinds of Wild Fruiting Body of Termitomyces

Abstract:
OBJECTIVE To compare and analyze the amino acid composition and medicinal value of three kinds of wild Termitomyces. METHODS The fruiting bodies of Termitomyces were hydrolyzed by HCl and determined by Hitachi L-8800 automatic amino acid analyzer. RESULTS The results showed that the contents of 18 kinds of total amino acids and essential amino acids and medicinal amino acids were 13.62%~25.53%, 6.50%~10.25% and 9.29%~16.64%, respectively. CONCLUSION Based on the results,it was concluded that the tested three kinds of Termitomyces had higher trophic and medicinal value and a vast developing prospective.
Determination of 6 Hazard Elements in Spatholobi Caulis by ICP-MS and Its Limit Assessment

Abstract:
OBJECTIVE To develop a method for the determination of 6 hazard elements such as Cr, Cu, As, Cd, Hg and Pb in Spatholobi Caulis. METHODS After the microwave digestion and then 6 hazard elements were determined by ICP-MS at the same time. Based on this, the limit of Cr, Cu, As, Cd, Hg and Pb were assessed with WHO standards. RESULTS The determination results of 6 hazard elements all have a good linear relationship, r≥0.9998. The limit of detection was range from 0.0016~0.0227ng/mL, the average recovery was 70.05%~117.98%, and RSD were 1.30%~3.69%. Determination results of samples of 6 hazard elements less than the maximum limit value(0.8000, 140.0, 0.8571, 0.4000, 0.2286, 1.429mg/kg). CONCLUSION The method is simple, accurate and high sensitive, can be used for the quality control and limit assessment of Spatholobi Caulis.
