Cyto-membrane Immobolized Chromatography:Hypothesis,Method and Application in Traditional Chinese Medicine
ZHU Quan
2006, 22(1)
Abstract:
A wide variety of molecules, including bormones, drugs, neurotransmitters, growth factors, antigens, viruses, plasma lipoproteins and glycoproteins, elicit their biological effects by virtue of their binding to cellular receptor. Many of these agents, however, interact with receptor or other target on external surface of the cell imbedded in cellular plasma membrane.Although in most circumstance,we still do not know correctly which substance are responsible for the effects of Traditional Chinese Medicine, and through which mechanism it works. But based the clinical response and pharmacological experimental phenomena,we believe that most Traditional Chinese Medicine's effect come from their compnents which interact with target of the organism. It is generally that noncovalent binding of drug to the target on the cell membrane may appear to be essentially reversible.Receptor and other target extend through the cell membrane. The part of the target on the exterior of the cell surface capture the signal components of TCM to be communicated to the cell in the outside environment. So our working hypothesis has been put forward as:The cell membrane of organism could be used as a material with which effective chemicals in Traditional Chinese Medicine could bind to the target that embedded on it.That is basis of cyto-membrane immobilized chromatography and it's application in Traditional Chinese Medicine. The cyto-membrane lmmoboliged chromatography is composed of the biologic membrane immobilized part and the chromatography part,The apavts have beon separatedin our wark, both parts have their optimal situation. This may be the creational in our work. The applications of red blood cell membrane immobilized chromatography in Angelica archangelica, platelet membrane immobilized chromatography in Salviae miltiorrhizae radix and Mailuoning Injection,a formula herbal injection for blood stasis, have been analyzed. The results shown that cyto-membrane immobilized chromatography are worked, although it is a pilot job.

Abstract:
OBJECTIVE To understand the effect and significance of extract of Cordyceps Sinensis on mice with viral myocarditis. METHOD 100 male BALB/c mice were randomly divided into a control group,an infection group and a group treated with extract of Cordyceps Sinensis. The mice in the infection group and the group treated with extract of Cordyceps Sinensis were inoculated with Coxsackie B3 virus and were then killed seven and fourteen days respectively after the inoculation.Myocardium SOD,MDA and iNOS expressions were determined and semiquantitative scoring of the pathological changes of the heart was carried out under the optical microscope. RESULT Compared with the infection group,SOD activity and iNOS expressions in the group treated with extract of Cordyceps Sinensis increased markedly(P<0.01),MDA level decreased(P<0.01) and myocarditis was relieved.CONCLUSION Extract of Cordyceps Sinensis can alleviate lipid peroxidation of mice myocardium by increasing active antioxidase and inducing iNOS expression.

Abstract:
OBJECTIVE To explore into the effect and mechanism of using As_2O_3 nanometer microballoon magnetism induced heating in the treatment of cervical carcinoma. METHOD Athymic mouse transplantation tumor models of human cervical carcinoma was made using SiHa cells.As_2O_3 nanometer microballoons, magnetic nanometer microballoons and As_2O_3 magnetic nanometer microballoons were then infused into the inoculation location of the tumor cells, which were then heated for 3 times with high-frequency alternating magnetic field. The experiment lasted for 45 days. RESULT Under the high-frequency alternating magnetic field,no tumor growth was detected in the As_2O_3 magnetic nanometer microballoon group, the tumor growth inhibiting rate being 99.93%,markedly higher than the As_2O_3 nanometer microballoon group(85.20%,P<0.01) and the magnetic nanometer microballoon group(87.72%,P<0.01).No marked differences were detected between the experimental groups and the control group in serum AST,ALT,Bun and Cr(P>0.05).No marked lesion was found in the tissue around the tumors and internal organs.CONCLUSION Under the high-frequency alternating magnetic field,As_2O_3 nanometer microballoons, magnetic nanometer microballoons and As_2O_3 magnetic nanometer microballoons are all effective in treating cervical carcinoma, especially As_2O_3 magnetic nanometer microballoons.As_2O_3 magnetic nanometer microballoons used in combination with fluid thermotherapy has dual anti-cancer effect.
