苍耳亭缓解过敏性皮炎及机制探索
Study on Mechanism of Xanthatin Alleviating Allergic Dermatitis

摘要: 目的 探讨苍耳亭对过敏性皮炎的药效作用及其机制。方法 利用异硫氰酸荧光素(FITC)诱导Th2型变应性接触性皮炎(ACD)模型观察苍耳亭全程给药对小鼠的耳肿胀、耳组织病理学以及脾脏指数的影响,并运用ELISA法检测小鼠耳匀浆中的Th2型炎症因子IL-4、IL-5和IL-13水平。利用MTT法考察不同化合物浓度对于人永生化角质形成细胞(HaCaT)、单核巨噬细胞RAW264.7以及小鼠脾脏淋巴细胞的增殖毒性作用。ELISA法检测Poly(I∶C)和TNF-α联合作用于上皮细胞产生胸腺基质淋巴细胞生成素(TSLP)的量以及脂多糖(LPS)诱导下RAW264.7产生TNF-α的含量。结果 苍耳亭40 mg/kg能显著抑制ACD小鼠耳肿胀,并可明显降低耳匀浆中Th2型细胞因子IL-4、IL-5水平,病理切片显示其可明显改善耳肿胀程度以及炎性细胞的浸润。体外实验中0.1 μmol/L苍耳亭对上皮细胞产生的TSLP有显著抑制作用;1 μmol/L的苍耳亭可以显著抑制RAW264.7产生TNF-α;苍耳亭对于脾淋巴细胞的增殖、对刀豆蛋白(ConA)以及脂多糖(LPS)诱导下T淋巴细胞和B淋巴细胞的活化增殖作用均未见明显影响。结论 苍耳亭对过敏性皮炎有明显的治疗作用,其机制可能通过抑制上皮细胞产生的TSLP以及巨噬细胞产生TNF-α发挥抗炎作用。Abstract: OBJECTIVE To investigate the effects of xanthatin on allergic dermatitis and its mechanism. METHODS Fluorescein isothiocyanate (FITC)-induced Th2 allergic contact dermatitis (ACD) was used and the effects of xanthatin on ear swelling, ear histopathology, spleen index and Th2 cytokines IL-4, IL-5 and IL-13 in mice ear homogenate were observed. The proliferation of xanthatin on HaCaT and RAW264.7 cells, and spleen lymphocytes were investigated by MTT assay. ELISA was used to detect the expression of thymic stromal lymphopoietin (TSLP) in epithelial cells induced by poly(I∶C) and TNF-α, and TNF-α in RAW264.7 induced by LPS. RESULTS 40 mg/kg xanthatin significantly inhibited ear swelling and reduced the production of Th2 cytokines IL-4 and IL-5 in ear homogenates in ACD mice. Ear pathology showed that xanthatin could improve the infiltration of inflammatory cells. In vitro, xanthatin obviously inhibited the production of TSLP in epithelial cells. Also, it markedly inhibited the production of TNF-α in RAW264.7 cells. While there was no significant effect on the activation of T lymphocytes and B lymphocytes induced by concanavalin A (ConA) and LPS. CONCLUSION Xanthatin shows obvious therapeutic effects on allergic dermatitis, and it may play an anti-inflammatory effect through inhibiting TSLP produced by epithelial cells and TNF-α produced by macrophages.
