      目的 探讨消癌解毒方对结肠癌的抗肿瘤作用及作用机制。方法 皮下注射结肠癌细胞CT26于BALB/c小鼠右腋下,建立荷瘤小鼠模型,连续灌胃给药14d后,处死小鼠,分离外周血单核细胞,流式细胞仪检测消癌解毒方对Treg细胞分布的影响,免疫组化检测消癌解毒方对肿瘤组织内Foxp3蛋白表达的影响。结果 消癌解毒方可以显著抑制结肠癌荷瘤的增长,降低CD4+Foxp3+Treg细胞在外周血及脾脏的分布,减少Foxp3蛋白在肿瘤组织内的表达。结论 消癌解毒方具有显著抑制结肠癌增殖的作用,其机制可能与减少外周血内Treg细胞的分布及肿瘤组织中Foxp3的表达进而影响其下游信号通路传导相关。
英文摘要:
      OBJECTIVE To explore the anti-tumor effect and mechanism of Xiaoai Jiedu Decoction on colon cancer. METHODS CT26 cells were injected subcutaneously into the right armpit of C57 mice to establish the tumor-bearing model. These mouse were sacrificed after 14d administration. Mononuclear cells of the peripheral blood were isolated and the distribution of Treg cells were detected by flow cytometry. The expression of Foxp3 protein in tumor tissue was detected by immunohistochemistry method. RESULTS Xiaoai Jiedu Decoction significantly inhibited the proliferation of colon cancer in mice, decreased the distribution of CD4 + Foxp3 + Treg cells in peripheral blood and decreased the expression of Foxp3 protein in tumor tissues. CONCLUSION Xiaoai Jiedu Decoction can significantly inhibit the proliferation of colon cancer by decreasing the distribution of Treg cells in peripheral blood and the expression of Foxp3 in the tumor tissues, thus affecting the downstream signal pathway.
