脾虚水湿不化证大鼠尿液代谢组学研究

Metabonomics Research on the Urine of Model of Dampness Stagnancy due to Spleen Deficiency Based on Liquid Mass Spectrometry

  • 摘要: 目的 以基于液质联用的代谢组学方法对脾虚水湿不化证模型及正常对照组大鼠的尿液进行研究,寻找模型大鼠尿液中差异代谢物,探讨脾虚水湿不化证的分子机制。方法 采用高脂低蛋白饲料加力竭游泳法复制脾虚水湿不化大鼠模型,收集模型组和正常对照组尿液,运用高效液相色谱-四极杆-时间飞行串联质谱仪技术检测2组尿液样本中的小分子代谢产物,对所得到的代谢指纹图谱进行主成分分析和偏最小二乘法-判别分析,找出具有差异的特征代谢物。结果 与正常对照组相比,模型组大鼠尿液差异代谢物为:α-酮戊二酸、瓜氨酸、乳酸、柠檬酸、5-羟色胺、谷氨酰胺等6种。结论 脾虚水湿不化模型大鼠机体糖、蛋白质代谢均出现紊乱,能量供应不足,胃肠功能下降,抗氧化能力下降,其机制可能与这些小分子代谢产物的变化有关。
