电针对中枢Stat5敲除所致肥胖小鼠瘦素水平和下丘脑Kctd15、Sh2b1 mRNA的影响

Effects of Electro-acupuncture on Serum Leptin Content and Expression of Kctd15 and Sh2b1 mRNA in Hypothalamus in Stat5-knockout Induced Obese Mice

  • 摘要: 目的 观察电针对中枢Stat5条件性敲除Stat5 Nestin-Cre (Stat5NKO)肥胖小鼠下丘脑Kctd15、Sh2b1基因表达的影响。方法 20只肥胖的Stat5NKO小鼠随机分为肥胖组和电针组,每组10只;10只Stat5fl/fl小鼠作为正常组。其中电针组选取单侧后三里、内庭穴进行针刺治疗,每日1次,双侧交替进行,每次30 min,韩氏电针仪频率2/15 Hz,疏密波,6 d为1个疗程,共治疗4个疗程。运用ELISA法检测血清中瘦素(Leptin)含量,qPCR技术检测下丘脑中肥胖相关基因Kctd15、Sh2b1 mRNA的表达。结果 与正常组小鼠比较,肥胖组小鼠体质量明显增加(P<0.01)伴随血清Leptin含量升高(P<0.05),下丘脑Leptin水平下降(P<0.05),同时下丘脑Kctd15、Sh2b1 mRNA表达下调(P<0.05~0.01);与肥胖组小鼠比较,电针组小鼠的体质量明显下降(P<0.01),血清Leptin含量降低(P<0.01),且下丘脑Leptin水平回升(P<0.01),Kctd15、Sh2b1 mRNA表达水平上调(P<0.01)。结论 电针可能通过调节Leptin水平并促进Kctd15、Sh2b1表达,实现对Stat5NKO小鼠的减肥效应。
