痛泻要方对溃疡性结肠炎大鼠结肠黏膜ERK1、ERK2基因表达的影响

Influence of Tongxie Yao Fang on ERK1 and ERK2 Gene Expressions of Colonic Mucosa in Rats with Ulcerative Colitis

  • 摘要: 目的 观察痛泻要方对溃疡性结肠炎(UC)模型大鼠结肠黏膜ERK1、ERK2基因表达的影响,探讨其作用机制。方法 将实验动物分为6组,采用TNBS/乙醇灌肠法造模,肉眼观察结肠大体形态损伤并进行评分,以大鼠结肠黏膜ERK1、ERK2为观察指标,采用RT-PCR法检测ERK1、ERK2基因表达水平。结果 肉眼观察模型组大鼠结肠组织黏膜层可见炎症和溃疡形成,与空白组比较,P<0.05,证实模型成功。模型组大鼠结肠组织ERK1、ERK2基因的相对表达量均高于空白组(P<0.05);治疗后,痛泻要方高剂量组、中剂量组ERK1、ERK2基因的相对表达量均较模型组升高(P<0.05,P<0.01)。结论 痛泻要方对TNBS/乙醇法UC大鼠模型结肠黏膜ERKl、ERK2基因的相对表达量有上调作用,提示痛泻要方治疗作用可能与ERK信号转导途径被激活有关。
