紫杉醇作用于肺癌A549细胞核转录因子荧光素酶值变化的意义

摘要: 目的 研究紫杉醇作用于肺癌A549细胞核转录因子荧光素酶值变化的意义。方法 于含10%胎牛血清RPMI1640培养基中,在37℃、5% CO2条件下细胞培养,转染核转录因子κB(NFκB)及环磷酸腺苷(CREB)质粒,单光子仪测定检测荧光素酶值,激光共聚焦显微镜观察NFκB核易位。结果 紫杉醇组NFκB及CREB荧光素酶均值分别为22387±1805、70403±4446明显高于未加药物的空白对照组1967±539、2607±665 (P<0.01),激光共聚焦显微镜观察到NFκB在紫杉醇作用下由细胞核转出到细胞质内。结论 紫杉醇作用于肺癌A549细胞后,核转录因子NFκB及CREB荧光素酶值升高,提示活性增强,紫杉醇可以使NFκB转入到细胞核。
